Objective1. To study the FT-IR and secondary structure of erythrocytes and the influence of arsenic on it,in order to provide an early indicator for arsenic poisoning damage;2. To research the concent of F-Hb and s CD163 in plasmal,so as to understand the damage of red cells and the state of inflammation;3. To make clear the damage effect of arsenic poisoning in liver tissue by the study of pathology;4. To research the expression change of CD68/C Dl63 in liver,in order to explore the impact of phagocytes and to provide the basis for the cell and molecular mechanism research of the early prevention and treatment in chronic arsenic poisoning;Methods1. Thirty-two male Sprague-Dawley rats were divided randomlyinto 4 groups:control group,80μg/L As treatment group,400μg / L As treatment group and 2000μg/L As treatment group.Animals were infected in sodium arsenite by the way of drinking water;2. Blood samples were obtained from abdominal aorta after As treatment,and the blood were be centrifuged and washed to get plasmal a nd the red cell suspension liquid;3. The sample of heart, liver, spleen, lung, kidney were obtained and the organ coefficients were recorded;4. Take a cetain suspension liquid drops to Ca F2,the samples weremeasured at room temperature by fourier transform infrared spectrometry(FT-IR); Analy the Amide I band through deconvolution and curve fitting to get the information of secondary structure in red blood cells;5. Erythrocyte parameters were measured with Sysmex Xt-2000 i automated blood analyzer;6. The level of FHb and s CD163 were detected by ELISA;7. The injury effect of hepatic pathology were detected by HE;8. Using immunohistochemical methods to analyze the change of CD68 and CD163 in rat liver.Results1.There was no significant difference in water intake,body weight and organ coefficient between infected group and control group;2.The samples’ spectrum absorption mainly includ Amide I, Amide II,C=C=N and-OH absorption peak;3.With the increase of the dose,the secondary structure of proteins in arsenic infected group was changed, and the high dose group is the most obvious(P<0.05);The percentage of the alpha helix is gradually reduced with dose increased;4.Each group of red blood c ell parameters have not yet occurred obviously abnormal change(P > 0.05);5.The content of FHb and s CD163 in arsenic infected group were obviously higher than that of control group, the difference was significant statistically(P<0.05),and there were certain relevance between FHb and s CD163(r=0.74);6.Liver tissue were significantly damaged after exposure, and there were some correlation between exposure concentration and damage degree;C D163 positive cell amount in arsenic infected decreased compared with that of control group,but the amout of CD68 positive cell was increased.Conclusion1. The relative content of alpha helix in red cells detected by FT-IR can be as an early indicator of arsenic poisoning damage;2. FHband s CD163 of Low levels of arsenic exposure can reflect the damage degree of red cell and the body’s level of inflammation;3. The number of CD68 positive macrophages which have a proinflammatory role were increased, and the number of CD163 positive macrophages which can ingest foreign particles were reduced,this may be ralated to the damage degree of liver in rats exposured to arsenic.
