Objective:This paper aims at studying the difference of cellular immunity function in different tumor cell proliferation state of advanced non-small cell lung cancer.Methods:For each of 89 naive advanced NSCLC patients from the first respiratory department of Shanxi Tumor Hospital during January 2014 and September 2015, the expression degree of proliferating cell nuclear antigen(ki-67) in cancer tissue was detected using Streptavidin-Perosidase immunohistochemical staining; Flow cytometry was used to measure the levels of cellular immune function, including the level of CD3+CD4+ helper T cells(Th), the level of CD3+CD8+ cytotoxic T cells(Tc), and the level of CD4+CD25high CD127 low regulatory cells(Treg) in peripheral blood; Cytometric beads array(CBA) was used to determine the levels of IFN-γ, IL-2 and TNF-α in peripheral blood serum. Using ki-67 immunohistochemical semi-quantitative method, the research subjects were divided into ki-67 positive group and ki-67 negative group to compare the immune cells and cytokines levels of reaction cell immune function between these two groups.Results:(1) The proportions of Treg in the positive expression of ki-67 were significant higher than those in negative subjects, that is(7.62±1.28)% vs(4.17±1.85)%, P<0.01 in detail, which is statistically significant; and the levels of Th, Tc, IFN-γ,IL-2 and TNF-αwere lower in the positive expression of ki-67 than those in negative subjects, which are(32.08±9.73)% vs(36.94±7.66)%, P < 0.05,(28.11±10.02)% vs(33.68±11.57)%, P <0.05,(8.07±1.64)pg/ml vs(11.41±2.60)pg/ml, P < 0.01,(6.22±2.84)pg/ml vs(8.11±4.92)pg/ml, P < 0.05, and(4.74±1.66)pg/ml vs(6.17±2.53)pg/ml, P < 0.01,respectively;(2) All the indexes, Treg, Th, Tc, IL-2, IFN-γ, and TNF-α, had few statistical differences in different clinical features advanced NSCLC patients;(3) The correlation coefficient between Th and Tc was-0.30 in the positive expression of ki-67, P<0.05. The correlation coefficient between IL-2 and TNF-α was0.75 in the positive expression of ki-67, P<0.01;The correlation coefficient between IL-2 and TNF-α was 0.93 in the negative expression of ki-67, P<0.01.Conclusions:The cellular immune function of advanced NSCLC patients reduced significantly with an increase in activity of tumor cell proliferation. The tumor cells proliferating actively break the cellular immune balance of normal cells by increasing the number of Treg and decreasing the levels of Th, IFN-γ, IL-2 and TNF-α, and the cytokines interact and interfere with each other, thus increasing the tumor progressing. The combined detection of tumor tissue expression level of ki-67 and the levels of Treg, Th, IL-2, IFN-γ and TNF-α in peripheral blood serum can provide certain reference value for the evaluating the individualized immunologic function of patients with advanced NSCLC.
