Objective:To investigate effects and possible mechanism of Intermedin(IMD) on tube-like structure formation of rat Renal Glomerular Endothelial Cells(rRGECs) in hypoxia. Methods:The rRGECs were randomly divided into three groups: normal group, hypoxia group and IMD group after culture in vitro. The latter two groups were placed in the hypoxia environment of 5% O2 incubating 6 hours. The normal group were incubated in normoxia. The viability of cells was examinated by MTT. The tube-like structure formation was detected by tube formation assay. The protein and mRNA express of VEGF and VE-cadherin were respectively measured by Immunocytochemistry and RT-PCR. Results:Compared with nomal group, the viability of cells, tube length, loops and branches of tube-like structures, and the protein and mRNA expression of VE-cadherin in hypoxia group all decreased(p<0.05), but those of VEGF in hypoxia group increased(p<0.05). Compared with hypoxia group, the viability of cells, tube length, loops and branches of tube-like structures, and the protein and mRNA expression of VE-cadherin in IMD group all increased(p<0.05). Conclusion:Intermedin can promote cell proliferation and tube-like structure formation, protect RGECs against hypoxia injury. The effect may be relactive with VEGF.
