Reactive Oxygen Species Induced Activation Of Myocardial Extracellular Matrix Metalloproteinases And Its Molecular

Objective: To investigate the effects of different concentrations of hydrogen peroxide on H9C2 myocardial matrix metalloproteinase activity. To inquiry hydrogen peroxide acting on myocardial matrix metalloproteinase activation of signaling pathways: JNK / EMMPRIN / MMPs.Methods:Establish H9C2 myocardial cell culture models,CO2 content of 5%, 37 the incubator ℃and cultured.Grouping:(1)normal control group(2)Hydrogen peroxide intervention(3)Hydrogen peroxide intervention and JNK inhibitors.Methods:Expression by Western Blot to detect myocardial intracellular protein content. Cell viability was detected by MTT assay.Results:When using H2O2 concentration of 200 umol / L to intervention 24 h,MMP-2 in the control group(1.0 ± 0.04) compare with 200 group(1.7 ± 0.04), P <0.05,show increased expression of MMP-2 protein. MMP-9 and MMP-2 being equal, the result is the same.MMP-2 in the control group(1.0 ± 0.04) vs JNK inhibitor group(0.7 ± 0.02), P <0.05, show MMP-2 with JNK inhibitor protein expression will be reduced after the intervention. EMMPRIN in the control group(1.0 ± 0.04) vs JNK inhibitor group(0.65 ± 0.02), P <0.05, show EMMPRIN expression will be reduced.Conclusion:When final concentration of hydrogen peroxide 200umol/L intervention H9C2 cardiomyocytes 24 h,protein expression of MMP-2, MMP-9 in the most obvious effect,protein expression of p-JNK、EMMPRIN were also increased.After using JNK inhibitors interfere H9C2 cardiomyocytes,protein expression of MMP-2、MMP-9、EMMPRIN、p-JNK were decreased.Experimental results demonstrate that the hydrogen peroxide can be used to regulate the activity of MMPs by JNK / EMMPRIN.
