Objective:(1).To explore the klotho protein in gastric epithelial cells(GECs) proliferation effect of migration;(2). To make a parcel of klotho protein can be used for endoscopic thermal polymerization gel and to evaluate material;(3). By animal experiment of polymer gel in the treatment of gastric ulcer; Methods:(1).Cell proliferation experiment(EDU) and cell migration(Transwell) : thelogarithmic students long GECs is divided into experimental group and control group in the two groups, experimental group to join klotho proteins, the control group without.(2)the production of polymer gel and evaluation :to beta cyclodextrin parcel klotho protein made from nanometer microspheres, with chitosan, the F- 127, glycerol and other materials into a temperature sensitive gel, nanometer microspheres with mixed gel, the material is made by electron microscope.(3) Gastric retention test: will only 8 C57 mice were divided into 2 groups, control group and experimental group, respectively with physiological saline, containing 1% methylene blue klotho protein gel, at 0, 30, 60, 120 minutes after the stomach, intensity of fluorescence detection by living fluorescence imaging system.(4)Animal experiment: 60 SD rats were randomly divided into 4 groups, respectively is saline group and blank group, omeprazole group, klotho protein gel. Each rat lavage 1 times a day, were given saline, blank gel, omeprazole, Klotho protein gel. In Each rat lavage 1 times a day, were given saline, blank gel, omeprazole, Klotho protein gel.In the treatment of,3,7 1 day each group randomly drawn 5 rats, ulcer areameasurement.Ulcer specimen section after HE staining and PAS staining, PNCA observe each ulcer healing. Results:(1).Cell experiment: klotho protein group increment rate is 2.3 times in the control group, cell migration amount is 3.6 times in the control group.(2). Electron microscopic observations: 50-200 nm in diameter nanometer microspheres, nanometer microspheres is evenly distributed on klotho gel.(3).Gastric retention test: klotho gel of fluorescence intensity in each time period were significantly higher than the control group.(4). Animal experiment: klotho gel group situation from the point of macroscopic view and histological healing is superior to the control group. Conclusion:(1) Under acid condition proved Klotho protein can promote proliferation of GECsmigration(2) Package klotho protein gel preparation can successfully used for endoscopic andadhesion in the gastric ulcer(3) Proved the klotho protein gel can promote gastric ulcer in angiogenesis, thus promotethe healing of ulcer area.
