Objective:To investigate the effect of PPAR-γ gene on proliferation,migration and invasion activity of human keloid fibroblasts(KFs).Methods:PPAR-γ gene was cloned into lentivirus vector,then the lentivirus particles expressing PPAR-γ were infected into KF cells.1.Real-time PCR and Western blot were performed to examine the expression of PPAR-γ in lentivirus infected cells.2.The growth ability was detected by MTT assay.3.The invasion and migration were detected by matrigel invasion assay and wound healing assay.Results:1.Comparing to controls group and KF-NC group,the expression levels of PPAR-γm RNA and protein were both significantly up-regulated after 96 hours of PPAR-γlentivims infection(P<0.05).2.Comparing with the KF-NC group and KF group,MTT assay showed that the A490 of KF-PPAR-γ group was down-regulated after lentivims infection(P<0.05).3.Transfection of PPAR-γ lentivirus inhibited the migration and invasion activity of KF cells(P<0.05).Conclusions:1.Lentivirus-mediated PPAR-γ gene efficiently inhibits proliferation,migration and invasion activity of KF cells.2.It suggests that improve the content and activity of PPAR-γ in early keloid, which may provide an effective way for the treatment of keloid.
