Objective:To study the influence of intervertebral foramen injection interferonα-2b on the analgesic effect of neuropathy pain model CCI rats and the TNF-α expression quantity of spinal cord intumescentia lumbalis.Methods:There are 48 healthy adult male SD rats, which are randomly divided into 4 groups(n= 4): sham operation group(S), which only expose the right sciatic nerve and have no ligation; CCI model group(CCI) : building CCI model, 7 days after model building, it conducts an intervertebral foramen injection of saline solution; Low-dose interferon α-2b CCI group(LCCI): building CCI model, 7 days after model building, it conducts an intervertebral foramen injection of 60,000 U interferon α-2b; High-dose interferon α-2b group(HCCI): building CCI model, 7 days after model building, it conducts an intervertebral foramen injection of 80,000 U interferonα-2b. It measures thermal pain threshold in the day before CCI processing(T0), one day after CCI processing(T1), three days after CCI processing(T2), seven days after CCI processing(T3) and one day after intervertebral foramen injection(T4), and three days after intervertebral foramen injection(T5). It randomly kills 4 rats in each group on the seventh day of measuring thermal pain threshold after CCI model building, the first and third day of intervertebral foramen injection, and takes the spinal cord of intumescentia lumbalis to make an Elisa detection.Results:1. At T0, there is no significant difference for basic thermal pain threshold of rats in each group(P > 0.05); At T1, T2 and T3 after CCI model building, compared with S group,the thermal pain threshold of group CCI, group LCCI and group HCCI has a statistical significance(P < 0.05), and three groups successfully build their CCI pain model; After an intervertebral foramen injection of saline solution or interferonα-2b, it measures the thermal pain threshold at T4 and T5, the thermal pain threshold of group CCI, group LCCI and group HCCI has no significant difference(P > 0.05), and it is prompted to have an intervertebral foramen injection of 60,000 units and 80,000 units of interferon α-2b and have an effect on thermal pain threshold of CCI model rats.2. After the successful CCI model building, at T3, CCI, compared with group S, the content of TNF-α in spinal cord of group CCI, group LCCI and group HCCI is statistically significant(P < 0.05), and it is suggested that TNF-α in spinal cord participated in pain formation process of CCI model and presented a high expression. After having an intervertebral foramen injection of saline solution or interferon α-2b, at T4 and T5,compared with the content of TNF-α in spinal cord, the content of TNF-α in spinal cord of group CCI, group LCCI and group HCCI is statistically significant(P < 0.05), and continued to have a high expression. The comparison among group CCI, group LCCI and group HCCI is non-significant(P > 0.05), and it is suggested that having an intervertebral foramen injection of 60,000 units and 80,000 units of interferonα-2b didn’t have a significance effect on the expression quantity of TNF-α in spinal cord. When conducting a comparison among groups, the comparison of group S at each time point is still not significant(P > 0.05); The comparison of expression quantity of TNF-α of Group CCI,group LCCI and group HCCI at T3 and T4 is more statistically significant(P < 0.05), and it is suggested that, between 7 to 10 days after CCI modeling, the expression quantity of TNF-α in spinal cord has a tendency to reduce.Conclusions:1. TNF-α in spinal cord participated in the development process of neurological pain CCI model, and presented a high expression. Between 7 to 10 days after CCI modeling, the expression quantity of TNF-α presented a downward trend.2.The study didn’t observe the relaxation effect of thermal pain threshold of using60,000 units and 80,000 units of interferon α-2b, but their effect on mechanical pain threshold cannot be ruled out.3. The study didn’t observe that using 60,000 units and 80,000 units of interferon α-2b has a significant effect on the expression quantity of TNF-α in the intumescentia lumbalis of CCI model rats.
