Objective:By researching the characteristic of Cy7-NYZL1 and the ability of marking bladder cancer xenografts in nude mice,lay the foundation for early diagnosis in future and precise treatment of bladder cancer.Methods:Determinating the most suitable concentration which made the fluorescent probe Cy7-NYZL1 has the strongest effect In vitro and establishing model of bladder cancer xenografts in nude mice.Nude mice would be injected Cy7-NYZL1 via the tail vein,after injecting,we would observe the distribution of Cy7-NYZL1 in nude mice through in vivo small animal imaging device.Results:When the excitation wavelength was 760 nm,As the concentration increases,absorbance of Cy7-NYZL1 increases.When the concentration was 300μmol/L,Cy7-NYZL1 has the highest absorbance.Then as the concentration increases,absorbance of Cy7-NYZL1 has no significant changes at a lower level.After injecting 200μL 300μmol/L Cy7-NYZL1 via the tail vein,Cy7-NYZL1 distributed throughout the body with the blood circulation and the fluorescence signal gradually increased.After 4 hour,the tumor’s absorbance achieve the highest.After that, the fluorescence signal gradually decline except tumor,After 12 hours,the fluorescence signal can’t be observed.Conclusions:The Cy7-NYZL1 fluorescent probe’s optimal concentration was 300μmol/L,the probe has specific targeting to BIU-87 bladder cancer,The optical imaging time window was about 4 hour after intravenous injection. |
