Objective To explore the effects of Chinese medicine tin caulis dendrobii on serum lipid level in mice, the female kunming mice were fed a high fat food, and then fed with different dose of aqueous extract of Dendrobium by gavage. and the content of mice serum total cholesterol(TC), triglycerides(TG) were detected. Furthermore, the, intestinal flora diversity of mice were analysis using IIIumina sequencing platforms to explore the influence of different doses of Dendrobium water extract on the, intestinal flora.Method 72 female mice were bought and adaptive fed for 3 days, then divided into 8 groups randomly: Normal diet group; Dendrobium low dose group; Dendrobium dose group; Dendrobium high dose group; high-fat diet group; Dendrobium fat diet and low dose group; Dendrobium fat diet dose group; and Dendrobium fat diet high dose group. Mice were daily regularly treated by gavage with 0.3ml aqueous extract of Dendrobium each time for each mouse for 2 weeks. After two weeks of treatments, the weight change, was evaluated and the TC,TG levels in the serums were detected. Furthermore, IIIumina sequencing platform was used to analyze the intestinal flora abundances and diversities.Results 1. After gavaging with low, medium and high dose of Dendrobium candidum water extract, the weights of the normal diet mice were increased. And there was significant difference between the high-dose group mice and normal diet group, thus verified the “Yiwei” effect of Dendrobium. Further the weight increased from the high-dose group mice and normal diet group, indicating that high-fat diet leaded to obesity in mice. 2. The high-fat diet group Kunming mice serum’s total cholesterol(TC) and triglyceride(TG) were significantly increase. Dendrobium water extract’s concentration is too high, so it had no effect on lipid metabolism disorder.3. The high-fat diet reduced the abundance and diversity of the mice intestinal flora. After the treatment of aqueous extract of Dendrobium gavage, the quantity and diversity of mice intestinal flora both increased. It can be found that the Dendrobium middle dose group and high dose group have similar intestinal flora abundance and diversity, Dendrobium low dose group similar to the high dose group of intestinal flor in caulis dendrobii structure, and the mice formed a relatively stable structure of intestinal flora after the water extract of Dendrobium gavage. It demonstrated that Dendrobium aqueous extract could improve mouse intestinal flora balance, and a relatively stable structure could be formed. 4.The structure of intestinal flora in mice was changed by high fat diet, and the dominant flora in the intestinal tract was changed at the same time. After 2 weeks treatment of high fat diet, the proportion of dominant bacteria in the intestinal tract of Kunming mice(Bacteroidetes) was increased, whereas the proportion of the thick wall bacteria(Firmicutes) was decreased. Dendrobium gavage of low, middle and high dose could significantly improve the abundance of Bacteroidetes in the intestinal tract. The expression of Firmicutes increased significantly in normal diet group mice when treated with high dose of intragastric administration of Dendrobium candidum water extract.
