Effect Of Peishishengxue Granule On Lymphocyte Proliferation Capacity And Serum IL–2 And IL–4 Levels In H22 Tumor-Bearing Mice

Objective: This study intends to use Peishishengxue granule intervention H22 tumor-bearing mice to have a better understanding of the influence and function mechanism in H22 tumor-bearing mice’s lymphocyte proliferation ability and the content of serum IL – 2 and IL – 4.Methods: Among 72 SPF Kunming mice, half male and half female, 12 mice were randomly selected according to their weight and sex to be the blank group. Considering the other 60 mice as a model which was established by subcutaneous inoculation with right anterior axillary region. After 24 hours of incubation, the mice were randomly divided into model group, Peishishengxue granule(PG) low dose group, Peishishengxue granule(PG) middle dose group, Peishishengxue granule(PG) high dose group, Zhenqi Fuzheng granule(ZG) control group. Feeding the male and female was at different cages. And medicines were administered to each group for 12 days by lavage. In addition, taking eyeball for blood in mice, the concentration of IL-2 and IL-4 in serum were detected by ELISA. After the mice were killed by the spinal dislocation method, stripped complete tumor tissue, thymus and spleen to weigh and record with electronic balance. The study uses MTT method to determine the proliferation ability of thymus and spleen lymphocytes. Tumor tissue sections, HE staining, observed the pathological changes of tumor cells.Results:(1)Compared with the blank group, the model group mice end with weight decreased significantly(P < 0.05); compared with the model group, the PG low,medium and high dose group and ZG group mice’s end with weight having different increases(P < 0.05); compared with ZG group, PG medium and high dose group end with weight increased significantly(P < 0.05).(2)The average tumor weight of that PG low, medium and high dose group and ZG group were lower than model group(P < 0.05).Tumor suppression rate of PG low, medium, high dose group and ZG group were 40.27%, 46.61%, 41.28%, 35.50%.(3) Compared with the blank group, the mice spleen index(SI) and thymus index(TI) of model group were significantly decreased(P < 0.05); compared with the model group, the PG low, medium and high dose group and ZG group’s mice spleen index(SI) increased(P < 0.05), the PG low,medium and high dose group and ZG group’s mice thymus index(TI) increased(P < 0.05). There were no significant statistic differences that the spleen index(SI) and thymus index(TI) of each PG dose groups compared with ZG group’s(P > 0.05).(4) Compared with the blank group, the mice thymocyte self proliferation, ConA induced proliferation capacity and proliferation rate of model group were significantly lower(P < 0.05); the mice spleen B lymphocyte self proliferation, LPS induced proliferation capacity and proliferation rate of model group were significantly lower(P < 0.05); the mice spleen T lymphocyte self proliferation, ConA induced proliferation capacity and proliferation rate of model group were significantly lower(P < 0.05). Compared with the model group, the mice thymocyte self proliferation, ConA induced proliferation capacity and proliferation rate of the PG low,medium and high dose group were significantly increased(P < 0.05); the mice spleen B lymphocyte self proliferation, LPS induced proliferation capacity and proliferation rate of the PG medium and high dose group were significantly increased(P < 0.05); the mice splenic T lymphocyte self proliferation, ConA induced proliferation capacity and proliferation rate of the PG medium and high dose group were significantly increased(P < 0.05). Compared with ZG group, the mice thymocyte self proliferation, ConA induced proliferation capacity and proliferation rate of PG medium dose group were increased significantly(P < 0.05); the mice spleen B lymphocyte self proliferation, LPS induced proliferation capacity and proliferation rate of the PG low,medium and high dose group were showed no significant difference(P > 0.05); the mice splenic T lymphocyte self proliferation, ConA induced proliferation capacity and proliferation rate of the PG low,medium and high dose group were showed no significant difference(P > 0.05).(5) Compared with the blank group, the mice serum IL-2 content of the model group was significantly lower(P < 0.05); compared with the model group, the mice serum IL-2 content of PG medium dose group was increased significantly(P < 0.05); compared with ZG group, the mice serum IL-2 content of the PG medium dose group was significantly increased(P < 0.05). Compared with the blank group, the mice serum IL-4 content of the model group was significantly increased(P < 0.05); compared with the model group, the mice serum IL-4 content of the PG medium, high dose group were decreased significantly(P < 0.05); compared with ZG group, the mice serum IL-4 content of the PG medium dose group was significantly lower(P < 0.05).(6)Slice observation of tumor tissue, Peishishengxue granule has inhibited the proliferation of tumor tissue in H22 tumor-bearing mice.Conclusion: Peishishengxue granule has the evident improvement to the H22 Tumor-bearing Mice’s body status, at the same time, there is a significant increase in lymphocyte proliferation and has a positive effect of the serum IL-2 and IL-4 content of H22 Tumor-bearing Mice.
