Object:To investigate the expression of human telomerase reverse transcriptase(hTERT) mRNA and PinX1 mRNA in deciduous cells from the bile of patients with cholangiocarcinoma, and evaluate its significance for clinical application. Methods:(1)Biles from 52 patients who were presented as “obstructive jaundice ” and operated percutaneous transhepatic cholangial drainage(PTCD) in stage Ⅰwere collected. 32 patients with cholangiocarcinoma(experimental group) and 20 corresponding patients with cholangiolithiasis(control group) were confirmed according to the result of postoperative pathological in stage Ⅱ.(2)The expression of hTERT and PinX1 mRNA in the deciduous cells of the bile of these two groups were investigated by reverse transcriptase-polymerase chain reaction(RT-PCR). Results:(1)hTERT mRNA was positive in 23 out of 32 bile samples of patients with cholangiocarcinoma(23/32,71.8%) than that in 3 out of 20 bile samples of patients with cholangiolithiasis(3/20,15%),and the expression level in experimental group were higher than that in control group(1.2046±0.76)vs(0.109265±0.267), which was significantly different between the two groups(P<0.05).(2) PinX1 mRNA was positive in 11 out of 32 bile samples of patients with cholangiocarcinoma than that 16 out of 20 bile samples with cholangiolithiasis, and the expression level in experimental group were lower than that in control group(0.212575 ± 0.30141)vs(0.677995 ± 0.35637), which was significantly different between the two groups(P<0.05).(3)Positive expression of hTERT mRNA were observed in 20 out of 20 bile samples of patients with lymphatic metastasis(20/20,100%) than that 3 out of 12 bile samples of patients without metastasis(3/12,25%), which was significantly different between the two groups(P<0.05). Positive expression of PinX1 mRNA were observed in 1 out of 20 bile samples of patients with lymphatic metastasis(1/20,5%) than that 10 out of 12 bile samples of patients without metastasis(10/12,83.3%),which was significantly different between the two groups(P<0.05).(4)The expression of hTERT mRNA and PinX1 mRNA in bile of patients with cholangiocarcinoma had a negative correlation. Conclusion:(1)The expression of hTERT m RNA in biles of patients with cholangiocarcinoma was significantly high while it was opposite about the expression of PinX1 mRNA, they were related to lymphatic metastasis, and this two gene had a negative correlation.(2)Combined detection may potentially serve as an important method to estimate the disease character of obstructive jaundice and make a great value for assessment of cholangiocarcinoma.
