Background: Malignant tumor has become one of the common diseases that threaten human health. According to a report released by the World Health Organization, we know that the incidence and death of lung cancer has became the first among all the malignant tumors, and still on the rise, so the prevention and treatment of lung cancer has became a widespread attention problem which throughout the world. At present, the treatment methods for lung cancer mainly include surgery, radiotherapy, chemotherapy, molecular targeted therapy, biological therapy, etc. But there are still many side effects, prone to drug resistance, the efficacy is not ideal, and so on. Traditional Chinese medicine, which is a traditional and unique treatment means in our country, it has unique effect in the prevention and treatment of lung cancer. The prescription used in this study is Yiqi Chutan formula(YQCTF) which has good curative effect for lung cancer. Long term clinical trials show that YQCTF is effective in the treatment of lung cancer.Objective: YQCTF used as the tool drug for intervention, human lung cancer A549 cells used as the target cells and BALB/C nude mice used as the object of drug action in this study. Tumor volume, tumor weight, and tumor suppression rate of lung cancer mice were measured and calculated, the expression of ATF6, XBP1 and CNX in transplanted tumors was detected. In this study, we demonstrated the specific anticancer mechanisms of YQCTF, and provided new ideas and direction for exploring the mechanism of traditional Chinese medicine in the treatment of tumor.Method: A549 lung cancer cells in logarithmic growth phase were inoculated in the back of the nude mice at the concentration of 1×107/mL. Every nude mice were inoculated 0.2mL. The mice were randomized into five groups, 8 in every group. including model group(saline group), cisplatin group(0.002g·kg-1), YQCTF low-dose group(3.0 g·kg-1), YQCTF high-dose group(6.0 g · kg-1) and combination group(6.0 g·kg-1/0.002g·kg-1). The mice of YQCTF groups and combination group were administered by gavag one time a day from 8th day after modeling for 14 consecutive days. The mice of cisplatin group and combination group were given by intraperitoneal injection one time a day from 17 th day after modeling for 5 consecutive days. On the 22 th day, all nude mice were euthanized. The tumor weight, tumor volume and the tumor inhibitory rate were detected. The expression of ATF6, XBP1 and CNX were detected with immunohistochemistry and quantitative real-time quantitative PCR and Western blotting.Results: The tumor weight and the tumor volume were lower in cisplatin group, YQCTF high-dose group and combination group than that in model group(P<0.01), and the tumor volume and weight of the combination group is lower than the cisplatin group(P<0.05, P<0.01). The mRNA and protein expression level of ATF6, XBP1 and CNX were lower in cisplatin group, YQCTF high-dose group and combination group than that in model group(P<0.01), and the average absorbance of ATF6, XBP1 and CNX were lower in cisplatin group, YQCTF high-dose group and combination group than that in model group(P<0.01). The average absorbance of ATF6, XBP1 and CNX were lower in combination group than that in cisplatin group(P<0.05, P<0.01). The mRNA and protein expression level of ATF6, XBP1 and CNX were lower in combination group than that in cisplatin group(P<0.01).Conclusion: YQCTF can inhibit growth of lung cancer. The mechanism is that it can decrease the expression of ATF6, XBP1 and CNX and reduce the unfolded protein response.
