Cellular immune response and humoral immune response can be induced by HIV infection, cellular immune control existing infections, the target of humoral immune is dissociative HIV. The classic vaccine strategy can not against HIV infection effectively. Another approach is to express HIV neutralizing antibodies in bodies by genetically virus vector.To construct recombinant vector, we use HIV neutralizing antibody gene 2G12 and the shuttle plasmid construct the dual promoter recombinant vector, which can express the light chain and heavy chain of the antibody. After identification of the recombinant virus. we package and amplify virus. To determinate titer and antibody expression. we infect cell with virus. To acquire the higher, longer antibody expression, we inject different virus carrier in mice.The HIV neutralizing antibody expression strategy of F2 A sequence was preliminarily studied. we cloned the heavy and light chains of neutralizing HIV antibodies separated by an F2 A self-processing peptide sequence into vector under the control of CMV promoter. 293 cells transfected with these vectors demonstrated secretion of human IgG into the culture supernatant could be detected. After identification of recombinant virus. we package and amplify virus. To determinate titer and antibody expression we infect cell with virus. To acquire the higher, longer antibody expression, we inject recombinant virus in mice.We inject several recombinant virus expressing antibody in mice, looking forward to a longer, higher expression of neutralizing antibodies in mice to achieve the goal of treatment for HIV/AIDS.
