Analyses Of Immune Microenvironment Papillary Thyroid Carcinoma With Hashimoto Thyroiditis

Objective: To investigate the immune microenvironment of papillary thyroid carcinoma(PTC) with Hashimoto thyroiditis(HT) and analyze its effect on the proliferation and development of papillary thyroid carcinoma. Methods: Tumor tissue of three cases papillary thyroid cancer with Hashimoto thyroiditis were harvested as an experiment group and tumor tissue of three cases papillary thyroid and three cases of Hashimoto thyroiditis were harvested as control group. PCR array which can profiles the expression of 84 key cytokines and chemokines crucial to the immune response was performed to analyze their effects on the proliferation and development of papillary thyroid carcinoma. Result: 1. The expression of 12 genes were upregulated significantly in PTC with HT compared to PTC(P<0.05): CXCL13、CXCL9、CCL19、LTA、CD40LG、IL16、IL24、IL7、BMP6、CXCL12、CCL22、TNFSF11,whereas the expression of 6 genes were downregulated significantly(P<0.05): CSF2、PPBP、CXCL2、IL8、LIF、BMP2. 2. The expression of 8 genes were upregulated significantly in PTC with HT, compared to HT(P<0.05): CXCL2、CCL17、IL6、CCL1、IL8、TGFB2、LIF、BMP4. 3. The expression of CXCL13 、 IL16、 CCL19 、 IL7 were upregulated significantly in HT(P<0.05) compared to PTC; whereas the expression of IL23A、CCL24、BMP2、LIF、PPBP、IL8、CXCL2 were downregulate significantly(P<0.05). Conclusions: 1. Differentiated cytokine and chemokine gene expression was detected in tumor tissue of PTC with HT, compared to PTC and HT. 2. Chemokine CCL19 and cytokine CD40 LG, IL24, LTA and TNFSF11 may suppress the development of PTC in tumor tissue of PTC with HT. 3. Chemokine CXCL2 and cytokine IL8, IL6 and LIF may promote to proliferation and development of PTC.
