Methods: 1.Cell morphological changes were observed under phase contrast microscope after treated by Gradient drugs at 48 h. 2.The MTT assay was used to detect the antiproliferative effect of FOD on BGC-823. 3. Flow cytometry was applied to determine the cell cycle and apoptosis of BGC-823. 4. Expression level of Ezrin and ICAM-1 was evaluated by Western-Blot. Results: 1. The phenomenon of cells volume reduced and attach decreased;the nucleus pycnosis, chromatin edge set;cell membrane integrity and somewhat less than 5-FU positive control groups, but com-pared with that in control groups there was significant variation. 2. Dealed with FOD for 24 and 48 h, the IC50 of BGC-823 cells was 12.58μg/mL and 8.43μg/mL,respectively. 3. The cell cycle of BGC-823 cells was blockaded in G0/G1 phase at the same time the apoptosis rate was remarkably increased compared with that in control group( p<0.05). 4. Expression of Ezrin decreased and ICAM-1 raised after dealed by gradient drugs at 48 h. Conclusion: 1. FOD could be in a dose and time-dependent mammer inhibit the proliferation and induce the apoptosis of BGC-823 cells. 2. It could significantly arrest cell cycle of BGC-823 cells at G0/G1 phase. 3. The low expression of Ezrin and high expression of ICAM-1 may contribute to the apoptosis Mechanism. | | Keywords/Search Tags: | FOD, cycle arrest, apoptosis-inducing, Ezrin, ICAM-1 | PDF Full Text Request | Related items |
