Objective:The long non-coding RNA(lncRNA) is a novel RNA that longer than200 nucleotides in length and lacking of protein-encoding capacity.Some researchers found that lnc RNA has key roles in the regulation of growth and development, especially to the proliferation and differentiation of stem cells. According to the analysis of differential expression of lncRNA after the osteogenic differentiation of hPDLSCs induced by Icariin, we selected the closely related lnc RNA and made a preliminary discussion on their function and meaning. This will provide new targets to the mechanism of differentiation of PDLSCs.Methods:(1) The hPDLSCs induced into osteogenic differentiation by ICA were used to detected by lnc RNA expression microarray, and the differentially expressed lnc RNAs and mRNAs were selected if the change of threshold values were >2.0 or <-2.0 folds and if P values were <0.05;(2) Bioinformatics analyzed differential expression of genes and predicted the target genes of osteogenesis related lncRNAs;(3) qRT-PCR was used to verify the results of expression microarray and detected the differential expression level of genes;(4) Combined with the above information, we preliminarily selected the target lnc RNA.Results:(1) During osteogenic differentiation, 48 lnc RNAs and 117 mRNAs were found to be differentially expressed(fold change >2.0 or <-2.0; P <0.05);(2)According to bioinformatics analysis, GO analysis showed that the most siginificant biological processes of the differently expressed genes, pathway analysis showed that the most significant pathways consisted of extracellular matrix, molecular and elastic fibers and the formation of elastic fibers, and the network reflected the interaction of mRNAs and lnc RNAs;(3) qRT-PCR verified 6 genes expression level and the trend was consistent with chip results;(4) lnc RNA-p16340 and lnc RNA-p21662 were chosen as the target molecules and lay a foundation for the study of the mechanism.Conclusions: Using gene microarray analysis and qRT-PCR method, we find the osteogenic differentiation related molecules lnc RNA-p16340 and lnc RNA-p21662.With the increase of induced time, their expression showed a rising trend. It may indicate that they are closely related to the regulation of osteogenic differentiation.
