Objective: Studies have found that Wdr5 shows very important effect involving stem cell differentiation, self-sustaining, and tumorgenesis. Wdr5 could promote urinary bladder cancer cells proliferation and self-renewal function via mediating H3K4 trimethylation. on the contrary, it can also inhibit osteoblast cancer cells proliferation by down-regulating Wdr5. Whereas there is no report on the role of Wdr5 in hepatogenesis yet. Our study aims on providing possible effects and mechanisms of Wdr5 on hepatocelluar carcinoma cells through generating Wdr5knock-out modles using lentiviral transfection method.Methods: We generated two shRNAs targeting Wdr5: shWdr5 A and shWdr5 B,and then infected Huh7 and HepG2 cells using lentiviral transfection. The expression level of Wdr5 was detected by qPCR and Western blot to indentify the transfection.MTT and clone forming assay were used to observe the effect of Wdr5 on cell proliferation. and the expression level of β-catenin was also tested to see if they are related to the possible mechanisms.Results: We constructed Wdr5 interfering cell models succesfully. The expression level of Wdr5 in Huh7 and HepG2 cells were significantly down regulated after tansfection. and the colony formation and cell proliferation were all inhibited after Wdr5 knockdown. and the expression level of β-catenin were all down-regulated after Wdr5 knockdown.Conclusions: Down regulating the expression of Wdr5 could inhibit hepatocellular carcinoma cells proliferation, and this could be involved in the regulation of β-catenin in Wnt/β-catenin signaling pathway.
