Objective:To investigate the influence of isoflurane on the expression of SIRT2 and RIP3 in hippocampal neuron in rats.Methods:Fifty-two Sprague-Dawley rats were randomly divided into 4 groups: control group(group C)received O2 for 6h, and isoflurane groups were received 0.75%(group I 0.75)、1.5%(group I1.5)、2%(group I2)isoflurane for 6h, respectively. After 12 h and 24 h the end of inhalation, brain hemisphere and the hippocampus were harvested, the location of SIRT2 and RIP3 were detected by using immunohistochemical staining, and the gene expression level of these two genes were quantified by real time PCR, respectively.Results:The location of SIRT2 and RIP3 were both found in the cytoplasm of hippocampus neuron. Compared with group C and group I0.75, the expression levels of SIRT2 and RIP3 increased in group I1.5 and I2 after the end of anesthesia for 12h(P<0.05). However there was no significant difference in the expression of SIRT2 in group 0.75% and 1.5% after the end of anesthesia for 24 h when compared to group C, while the expression level in SIRT2 decreased in group I2(P<0.05). There were no significant differences in the expression level of RIP3 among the four groups after the end anesthesia for 24 h. The expression level of SIRT2 and RIP3 increased after the end of anesthesia for 12 h,but decreased after the end of anesthesia for 24h(P<0.05).Conclusion:The expression level of SIRT2 and RIP3 are up-regulated by inhalation of 1.5% and 2% isoflurane for 6h after 12 h,but the level of SIRT2 is down-regulated by inhalation of 2% isoflurane after 24 h.
