Objective:To investigate the biological safety in vivo and osteogenesis of 3D printing of β-TCP scaffold loaded with sustained-release microspheres of Isoniazid and Rifampin/PLGA.Method:We carried out aseries of experimental studies on 3D printing of β-TCP(400μm) scaffold loaded with sustained-release microspheres of Isoniazid and Rifampin, the compound materials of 3D printing of β-TCP scaffold loaded with sustained-release microspheres of Isoniazid and Rifampin/PLGA were implanted in the femoral condyle of New Zealand rabbits and served as the loaded group,the rabbits implanted in scaffold without Isoniazid or Rifampin/PLGA were served as a unloaded group, and rabbits only received surgery were served as control group.X-ray observation, CT and Micro-CT scanning were performed at the 4th, 12 th and 20 th weeks, bone tissue were analyzed at the 20 th week to verify the osteogenesis and bone repair of scaffolds, degradation of compound materials.Venous bloods of rabbits in 3 groups were collected at the different time points of4 th, 12 th and 20 th weeks randomly, take heart,liver,spleen,lungs,kidney organization of the loaded group at the 20 th,and the liver and kidney of rabbits in the loaded group were obtained for comparison of relative parameters and tissue sections.Result:It was confirmed that the compound materials of 3D printingof β-TCP scaffold loaded with sustained-release microspheres of Isoniazid and Rifampin/PLGApresentedosteogenesisfunction with totally biological safety.Conclusion: The compound materials of 3D printing of β-TCP scaffold loaded with sustained-release microspheres of Isoniazid and Rifampin is ideal in the repair of tuberculosis bone defect, and it may be applied aspromisingclinical treatment of bone tuberculosis.
