| Objective: To explore the expression and the role of LZTS2 gene in gastric cancer and gastric stump carcinoma.Methods: Using immunohistochemical technology to detecting the expressions of LZTS2 and β-catenin in gastric cancer, remnant gastric cancer and normal gastric mucose tissue, and collecting the patients clinical data including age, gender, tumor differentiation, TNM staging,Lauren type, lymph nodes, the recurrence time of gastric stump carcinoma.The correlation between the expression of LZTS2 and β-catenin and the clinical data was comparative analysised.The plasmid containing LZTS2 gene was transfected into SGC-7901 cells.The cells were divided into transfection group, negative control group and blank control group. The expression of LZTS2 and β-catenin of cells in each groups were assayed by RT-PCR and Western blot.The ability of cell proliferation was detected by MTT in each group and the cell migration ability was detected by Transwell.Results: LZTS2 protein was expressed in the cytoplasm of normal gastric mucosa, the positive rate was significantly higher than that in gastric cancer tissues and gastric remnant cancer tissues(P<0.01). The abnormal expression of LZTS2 was not associated with the gender and age of patients, and was closely associated with the differentiation, TNM stage, Lauren’s classification,tumor size and lymph node metastasis. The positive expression of LZTS2 is negatively correlated with the abnormal expression of β-catenin. The transfection pCMV6-LZTS2 plasmid can significantly increase the LZTS2 protein expression, the difference was significantly significant between the experimental group and negative control group. The expression of LZTS2 mRNA of expremental group was significantly higher than that of negative control group, the difference was significantly significant. The expression of β-catenin was inhibited in the experimental group which was lower than that in the negative control group, the difference was significantly significant. The ability of proliferation was detected by MTT and that of experimental group significantly decreased. The migration ability of experimental group,detected by Transwell,decreased significantly.Conclusion: The expression of LZTS2 in gastric cancer and gastric stump cancer tissue was significantly lower than normal gastric mucose tissue, and may be associated with the occurrence,development, invasion and metastasis. LZTS2 can inhibit the expression of β-catenin and cell proliferation and migration in SGC-7901. |
