Objective:To observe the effect of betulinic acid(BA) on proliferation, apoptosis,migration in human colonic carcinoma cell HT-29,and to explore the mechanism of betulinic acid against tumor invasion and metastasis.Methods:HT-29 was treated by different concentrations of drugs(BA). The effects of cell proliferation, apoptosis,migration were assayed by MTT,FCM and Scratch Method respectively.The expression of Hpa mRNA was assay by real-time PCR.Result:1.The vitality of cells in drug intervention group was inhibited obviously in the morphology.2.The HT-29 was treated by different concentrations of drugs(5,10,20,40 ug / L)respectively. After 24 hours, the inhibition rates of proliferation were 13.5%, 26.1% and45.8%, 51.3%. After 48 hours, were 14.4%, 30%, 51.1%, 62.4%. After 72 hours, were19%, 35%, 53.8%, 65%.(P < 0.05)3.The apoptosis rates of HT-29 were treated respectively by different concentrations of BA for 48 hours was 3.4%, 9.2%, 20.4%, 27.4%, the apoptosis rate of control group(0ug/L) was 2.0%.(P<0.05)4.The migration distance of drug group was 144.21,127.14,101.52,73.32,respectively.And the non-drug group was 152.17.(um)5.The expression of Hpa mRNA in different group(0,5,10,20,40ug/L),were 1.000,0.910, 0.856, 0.694, 0.474.(p<0.05)Conclusions:BA can inhibit the proliferation, migration, and promote apoptosis of human colonic carcinoma cell HT-29,and BA may inhibit the invasion and metastasis of tumor by decreasing the expression of heparanase.
