The hsa-miR-29c levels of cancer tissues and adjacent tissues were detected by real-time PCR. All of subjects were followed up for more than three years. The expressions of hsa-miR-29c were detected using real-time PCR in ESCC cell lines (Eca109 and Kyse150), colorectal cell line HT-29, HepG2 cells and normal esophageal epithelial cell line (Het-la). Hsa-miR-29c overexpression were performed by transfection technology. The apoptosis and proliferation of cell lines overexpressing hsa-miR-29c were measured by flow cytometry and MTT assay.[Results]1. The expression of hsa-miR-29c in cancer tissues of ESCC patients were significantly lower than that in adjacent tissues (P<0.05);2. Compared with normal esophageal epithelial cell line Het-la, hsa-miR-29c expression in ESCC cell lines Eca109, Kyse150 and colorectal cell line HT-29, hepatoma cell line HepG2 were also significantly lower (P<0.01);3. Hsa-miR-29c expression in ESCC were correlated with lymph node metastasis and tumor stage(P<0.05);4. Downregulated hsa-miR-29c levels were also related with a shorter time to progress (TTP) (P<0.005);5. Overexpression of hsa-miR-29c can inhibit cell proliferation and promote apoptosis of ESCC cells.[Conclusion]Hsa-miR-29c showed low expression in esophageal squamous cell carcinoma, which may affect the occurrence and development of cancer by regulating cell proliferation and apoptosis pathway. | | Keywords/Search Tags: | ESCC, hsa-miR-29c, realtime PCR, regulation mechanism | PDF Full Text Request | Related items |
