Objective:This study is to explore the effects of transplantation of adipose derived stem cells combined with adiponectin on improving erectile function of in intraperitoneal injection of streptozocin(STZ) with high-fat diet induced type 2 diabetic rats.Method:Male Sprague Dawley rats were fed either with hight-fat diet combined with single small doses of STZ to induce type 2 diabetes or with basic feed as controls. All rats were randomly assigned into four groups:the normal control group(NC);diabetes group without any therapy(DM);the diabetes group treated with ADSCs(ADSCs);and the diabetes group treated with ADSCs combined with adiponectin(ADSCs-Adiponectin).Rat ADSCs were harvested and transplanted into corporal cavernosum of STZ with high-fat diet induced diabetic rats 8 weeks after extablishment of diabetes,besides,the rats of group ADSCs-Adiponectin were also treated with intraperitoneal injection of recombinant adiponectin.Four weeks after ADSCs transplantation,all rats were analyzed for erectile function by the ratio between intracavernous pressure(ICP) and mean arterial pressure(MAP) during electrostimulation of cavernous nerve.Masson’s trichrome staining was used to evaluate the fibrosis in the corpus cavernosum. Smooth muscle,endothelium and neurofilament in corpora cavernosum were assessed using immunofluorescence assay.Result:Four weeks after adipose derived stem cells transplantation combined with adiponectin,the ICP/MAP ratio of the group of ADSCs and ADSCs-Aadiponectin was increased significantly compared with diabetic control group(P<0.05).The fibrosis in the ADSCs combined with recombinant adiponectin treated rats and the group of ADSCs was significantly decreased compared with diabetic control group(P<0.05).The content of smooth muscle,endothelium and neurofilament in corporal cavernousa of the rats of group ADSCs-Aadiponectin and ADSCs was significantly increased compared to diabetic control (P<0.05).Conclusion:Intracavernous transplantation of ADSCs had beneficial effects on improving erectile function of diabetic rats.Moreover,Intracavernous transplantation of ADSCs combined with intraperitoneal injection of STZ had beneficail effects on improving erectile function of diabetic rats and increasing the content of smooth muscle,endothelium and neurofilament in corporal cavernousum.
