Objective: To explore the impact of pulsed electric fields with cell cycle specific agents paclitaxel on the human cervical carcinoma Hela cell proliferation,invasion and metastasis.Methods: Cervical cancer cell line Hela was cultured in vitro. Application of different concentration of the G2/M cell cycle specific agents paclitaxel or cell cycle nonspecific agents cisplatin with μs PEF of different electric intensity in cervical cancer Hela cell line. Drugs were administered at the pulse electric field within 6h and after 6h, respectively. Cell Proliferation was evaluated by CCK-8 assay; the invasion and immigration capacity was evaluated with transwell cave method.Results:(1)Different concentration of paclitaxel or cisplatin with μs PEF of different electric intensity can inhibit the Hela cell. Under the condition of low field intensity parameter has the concentration dependence; under the condition of low concentration of has the electric field intensity dependence(P < 0.05).(2)After being treated with pulsed electric fields with cell cycle agents, the invasion and immigration capacity of Hela cells were also inhibited.When Hela cells were treated with high intensity pulsed electric field(2.5k V/cm) after 6h, Invasion and metastasis of paclitaxel on Hela inhibition ability were better than cisplatin(P < 0.05).(3)High intensity pulsed electric field(2.5 k V/cm)with paclitaxel after 6 h on Hela cells, the inhibition of Hela cells viability enhanced than within 6h(P <0.05).Conclusion: Pulsed Electric Fields combining with paclitaxel or cisplatin on Hela cells, the ability of migration and invasion of cervical cancer Hela cells are significantly restricted, and when high intensity pulsed electric field acting on He La cells after 6h, the G2/M cell cycle specific agents paclitaxel on Hela inhibitory effect is superior to cell cycle nonspecific agents cisplatin. |
