Objective: To observe the effect of Roux-en-Y gastric bypass(RYGB)on the expression of glucagons-like peptide-1(GLP-1) in the plasma of rats diseased with spontaneous 2-type diabetes(GK rats)and glucose transporter-4(GLUT-4) of the skeletal muscle cell, to discuss its mechanism for improving the insulin resistance..Methods: 30 male GK rats were selected and randomly divided into three groups, respectively, surgery group, sham surgery group and diet controlling group,10 rats for each group. Meanwhile, 10 Wistar rats from the animal experiment center of our hospital were selected for a control group. The surgery group was performed with RYGB. The sham surgery group was only performed with open surgery for probe instead of real surgery. The rats in the diet controlling group were not provided with surgery and medication and were given a mere control over diet and exercise. The control group was left alone without any treatment. The level of GLP-1 in plasma of rats in each group before the surgery and at week 4 after the surgery had been detected and compared, 4 weeks later, the rats were killed and their rear legs’ skeletal muscle were taken for detection. The expression of m RNA of GLUT-4 in the skeletal muscle tissue of rats was detected with RT-PCR experimental method.Results: the GLP-1 of rats in the surgery group, sham surgery group, diet controlling group and control group at week 4 after surgery were respectively, 30.42±5.33 umol/L、6.98±0.32 umol/L、7.33±0.65umol/L and 29.65±3.76 umol/L, the preoperative level of GLP-1 of surgery group was 6.98±0.73 umol/L. for the surgery group, there was a significant difference between the level at week 4 after surgery and the one before surgery, P<0.05. At the time point of week 4 after surgery, there was a significant difference between the surgery group and sham surgery group and diet controlling group, P<0.05. The expression of m RNA of GLUT-4 of rats in the surgery group, sham surgery group, diet controlling group and control group at week 4 after surgery were respectively, 1.81±0.11、1.19±0.14、1.18±0.08 and 1.18±0.08, At the time point of week 4 after surgery, there was a significant difference between the surgery group and sham surgery group and diet controlling group, P<0.05. At the time point of week 4 after surgery,the difference in m RNA of GLP-1 and GLUT-4 of rats between the surgery group and control group was not statistically significant, P>0.05.Conclusion: 1 The level of GLP-1 in plasma of GK rats was significantly increased after roux-en-y gastric bypass surgery.2 the expression of GLUT-4m RNA in skeletal muscle of GK rats after roux-en-y gastric bypass s was increased, and the symptoms of insulin resistance were relieved.
