Objective:to observe the influence of liver PDX1 and FOXO1 expression in nonobese diabetic mice after interfered by anti-CD20 monoclonal antibody combined with interleukin10.Methods:Twenty-four female NOD mice were randomly divided into four groups.Mouse A:Anti-CD20, B:Anti-CD20+IL-10,.C:IL-10 and D:Normal Control(NC) groups were caudal venously injected of 250 ug Anti-CD20 antibody, combined Anti-CD20 antibody 250ug and 0.1ml IL-10,0.1ml IL-10 and 0.1ml physiological saline,respectively. But the first day,A and B groups were injected Anti-CD20 antibody 500 ug.All mice were monitored for blood glycose every day, and sacrified 6weeks after injection. Their serum levels of C-Peptide,TNF-α,IL-10,CD20 were measured by ELISA.The liver PDX1 and FOXO1 expressions were observed by immunohistochemistry、western blot and RT-PCR.Results: After combined Anti-CD20 and IL-10 intervention, the C-Peptide in the serum were enhanced(P<0.05) and the serum TNF-α were lower compared with other 3groups(P<0.05). The liver PDX1 expression was upregulated(P<0.05) and the the expression of FOXO1 in the liver was decreased(P<0.05).Conclusion:The combined administration of Anti-CD20 and IL-10 increased the expression of PDX1, and decreased the the expression of FOXO1 in the liver.
