Objective To investigate the expression of connection tissue growth factor(CTGF) in ovarian cancer tissues and cell lines,when the CTGF genes were blocked,the biological characteristics of ovarian cancer cell lines were explored.Methods 1. The expression levels of CTGF were examined by RT-PCR and immunohistochemical staining in ovarian cancers、benign ovarian tumors、normal ovarian tissues. 2. The expression of CTGF in human ovarian cancer cell lines,likes BG-1、SKVO3、A2780,were measured by western blot and RT-PCR,which can screen the relatively high expression of CTGF cell line. 3.The relatively high expression of CTGF cells were transfected with human pecific small molecule RNA CTGF genes, the expression level of interference effect detection in m RNA and protein expression were screened successfully silenced CTGF cells 4. Inspected the show of CTGF after transfectded by RT-PCR and western blot, analysis and detected the change of proliferation and invasion by MTT and Transwell.Results 1.The expression levels of CTGF in ovarian cancers were lower than that in Normal ovaries and benign ovarian tumors tissues by immunohistochemical staining and RT-PCR.χ2 analysis was used to show that the expression of CTGF was related to clinical stages、lymph node metastasis(P<0.05),but not age,Histologic type,Histologic grade,Peritoneal cytology(P>0.05) 2.BG-1 cell lines expresses high levels CTGF,evaluated by PCR and western blot. 3.CTGF si RNA was transfected to BG-1 cells, the expression of CTGF protein and m RNA in the experimental group compared with the control group, the expression decreased significantly, for the next step of the experiment selected successfully silenced celllines. 4.We surveyed that when CTGF genes were silenced,it enhances the ovarian cancer cells the ability to proliferate and invasion.by MTT and transwell.Conclusion The CTGF was lower expressed in ovarian cancers,χ2 analysis was used to show that the expression of CTGF was related to clinical stages、lymph node、metastasis,but not age,Histologic type,Histologic grade,Peritoneal cytology,CTGF silencing in ovarian cancer BG-1 cells by using the si RNA target, the and invasion of ovarian cancer cells were increased. The results suggest that CTGF plays an important role in the proliferation and invasion of BG-1 cell line, which can provide a new target for the treatment of ovarian cancer.
