Objective To investigate the frequency distribution of PPAR-δ-87T/C gene polymorphism in the gestational diabetes mellitus(GDM) and healthy pregnant women of Chinese Han population from Qingdao region, analyse the correlation between different genotypes and clinical indicators, and clarify the correlation between different PPAR-δ-87T/C gene polymorphisms and GDM in Han population from Qingdao region.Methods Use the case-control study. 150 women with GDM, who delivered through cesarean section at the Affiliated Hospital of Qingdao University, were selected as the GDM group, and another 160 normal pregnant women were chosen as control. The peripheral venous blood with mothers were collected. The serum levels of fasting plasma glucose(FPG), glycosylated hemoglobin(Hb A1c), total cholesterol(TC), triglyceride(TG), high density lipoprotein(HDL), low density lipoprotein(LDL) and fasting insulin(FINS) were measured in the women of both groups. Insulin resistance index(HOMA-IR) was calculated. Then DNA were extracted. Use the PCR technology to analyse PPAR-δ-87T/C gene, then further select some representative samples to take the sequencing test in order to verify the results of digestion.Results 1 Tested by Hardy-Weinberg equilibrium, the frequency distribution of TT genotype, CT genotype, CC genotype, T allele, C allele in GDM group and control group was consistent with genetic equilibrium. And people selected had group representative. 2 In GDM group and control group, there were no significant differences in the frequency distribution of genotypes and alleles of PPAR-δ-87T/C(P>0.05). 3 In GDM group, TG level of CC genotype was significantly higher than TG level of TT genotype(P<0.05). However, there were no significant differences in level of age, prenatal BMI, postpartum BMI, FPG, Hb A1 c, TC, HDL, LDL, FINS and HOMA-IR(P>0.05). 4 In control group, there were no significant differences in level of age, prenatal BMI, postpartum BMI, FPG, Hb A1 c, TC, TG, HDL, LDL, FINS and HOMA-IR(P>0.05).Conclusion There is no significant correlation between PPAR-δ-87T/C gene polymorphism and the incidence of GDM in the Han population of Qingdao, but the PPAR-δ-87T/C gene polymorphism may be related to the disorder of lipid metabolism in GDM.
