The Effects And Mechanisms Of Apoptosis In Human Ovarian Cancer Cells Treated With Huaier Particles

Objective: To explore the effects and mechanisms of Huaier particles on proliferation and apoptosis in SKOV-3 cells.Methods:Morphological changes were detected by phase contrast microscope in SKOV-3cells treated with various concentration of Huaier particles for 48 h. The ability of SKOV-3cells was measured using CCK8 assay after incubation with different concentration of Huaier particles for 24 h, 48 h and 72 h respectively. Apoptosis and cells cycle was analyzed by FCM in SKOV-3 cells treated with various concentration of Huaier particles. Bcl-2, Bax,cleaved-caspase3 and Cyt c protein were detected by Western blot in SKOV-3 cells after treatment of various concentration of Huaier particles. Effects of Huaier on mitochondrial membrane potential in SKOV-3 cells was measured by JC-1 assay.Results:Morphological changes of SKOV-3 cells exposed to Huaier particles for 48 h were detected by phase contrast microscope. Huaier particles inhibited the ability of SKOV-3 cells.With the increase of drug concentration, inhibition enhanced gradually in dose and time-dependent manner. The results of cell cycle indicated that cell cycle of SKOV-3 cells exposed to Huaier particles for 48 h was retarded in G2/M. Huaier particles significantly reduced apoptosis and increased the early apoptosis in SKOV-3 cell for 48 h in dose-dependant manner. Western blot analysis showed that Bax, cleaved-caspase3 and Cyt c were up-regulated and Bcl-2 was down-regulated in groups treated with Huaier particles.Huaier particles induced the loss of mitochondrial membrane potential(MMP) in SKOV-3cells.Conclusion : Huaier particles can significantly suppress the ability and also induce the apoptosis of human ovarian cancer SKOV-3 cells in vitro. The mechanism of Huaier particles inducing apoptosis of SKOV-3 cells might be related to the activation of mitochondria-dependent apoptosis signaling.
