Surgical treatment is the first choice for clinical treatment of breast cancer. Radiotherapy and chemotherapy are the auxiliary means for breast cancer treatment. However, because of the obvious toxic side effects of cancer chemotherapy, tumor chemotherapy has a strong systemic toxicity, so it is urgent to explore new therapeutic method.Integrin receptor is a receptor expressed in the surface of tumor cells. It is common in breast cancer, glioma, liver cancer and melanoma, but less on other normal cells, so the integrin receptor has become a hot research topic in tumor targeting therapy. Some studies have shown that RGD is a specific ligand for integrin receptors, and RGD peptides have high efficiency in the selection of integrin receptors. It can, through contact with the cell membrane and inside the cell, is not selective.In this study, the R8-RGD modified doxorubicin liposome was prepared by using adriamycin as a model drug, which is commonly used in tumor chemotherapy. By covalent binding, R8-RGD is attached to the surface of liposomes, and the R8-RGD modified doxorubicin liposome is prepared.The first part of this paper is the cultivation of MCF-7 cells and the experimental group. MCF-7 cells were cultured in the laboratory, and the cells were cultured and divided into groups. The MCF-7 cells were randomly divided into PBS group, group, RGD modified doxorubicin liposome group, R8 modified doxorubicin liposome group and RGD-R8 modified.In the second part of this article, we will prepare different adriamycin liposome, investigate its physical and chemical properties, and evaluate the cell level. First, the characterization of RGD-R8 modified doxorubicin liposome was investigated. The results showed that the prepared RGD-R8 modified doxorubicin liposome was spherical and the particle size was uniform. Because of the red fluorescence of adriamycin. We detected the uptake of MCF-7 cells in breast cancer cells with adriamycin, the results showed that the uptake efficiency of RGD-R8 cells in breast cancer MCF-7 cells was significantly higher than that in other groups, the difference was statistically significant(P<0.01). We further investigated the mechanism of MCF-7 cell uptake by RGD-R8 modified doxorubicin liposomes. The results show that the uptake of RGD-R8 modified doxorubicin liposomes by MCF-7 cells is a temperature, energy and charge dependent uptake process by the grid proteins. After we carried out the cytotoxicity test, the results showed that the inhibition effect of RGD-R8 on the tumor cells was significantly stronger than that of other liposomes, which was the same as that of the cells.To sum up, this study successfully prepared RGD-R8 modified doxorubicin liposome, which has the potential of breast cancer cell targeting and breast cancer cell growth inhibition, and is a promising drug delivery system for the treatment of breast cancer.
