Choline Acetyltransferase Expressed By Radial Glial From Rat Fetal Brain

To determine whether radial glias can generate cortial cholinergic neurons,the choline acetyltransferase expression of radial glias in the dorsal telencephalon was approved.To observe the morphology,mitosis and the expression of Choline acetyltransferase of radial glias which generate the cortical pyramidal neurons in developing dorsal telencephalon, immunohistochemistry staining,electronic microscopy techoniques were used.First of all,we immuno-labeled the Radial Glia with the monoclonal antibody Vimentin in the cell of culture telencephalon(Vimentin is the special marker of Radial Glia).The result showed that the bodies of Vimentin positive cells located in the ventricular zone.secondly the telencephalons cell of rats at embryonic 14 days(E14) were primary culture,then Immunocytochemical double staining for vimentinand choline acetyltrans-ferase to test whether ChAT was expressed in the population of radial glial cell in vitro.The result indicated that there were primary culture,then Immunocytochemical double staining for vimentinand choline acetyltrans-ferase confirm E14 rat telencephal brain cells cultured in vitro with cells Vimentin/ChAT Immunochemistry double staining positive expression.In vivo and in vitro experiments common confirmed that in the E14 rat telencephal brain ChAT was expressed in the population of radial glial cell.It is suggested that the cell fate of the population of radial glial cell has been decided in the asymmetrical cell division,and then synthesizes into neuroteansmitter acetyleholine. Providing a theoretical basis for further study cholinergic neurons in the development of mechanisms(1) Radial Glia can generate cholinergic neurons;(2) it is possible that Radial Glia can be classified into various sub-groups according to generating different kinds of blastneurons;(3) The Radial Glia may act the important role in the developing cerebral cortex.The radial glias of doral telencephalon can generate cortical cholinergic neurons.
