We had developed an ethanol injection method, one of the simplest methods, to obtain large unilamellar vesicle liposomes of curcumol. Through orthogonal experimental design, one optimum recipe of curcumol liposomes was founded. A simple preparation method-ethanol injection method was modified to achieve preferable fidelity and high percentage envelopment.A method was developed for the determination of envelopment and curcumol concentration in liposomes and biological samples by HPLC; it was proved that this method was accurate, sensitive, specific and good enough to be used in pharnacokinetic and tissue distribution study of curcumol.The chemicophysieal properties and stability of curcumol liposomes were studied. The large unilamellar vesicle liposomes were seen though electron microscope. The mean diameter and Zeta potential of liposomal were 118 nm and -31.7 mV, respectively. In accelerated tests, which contained high temperature, low temperature and light exposing experiments, it was shown that liposome injection was more stable than water solution. The liposomal injection had little change after it was stored at 4℃for 6 months. Due to the oxidization and hydrolysis of phospholipids, it was advised that the production should be stored at 4℃in oxygen-free atmosphere from light.After intravenous injection of curcumol water solution and liposomes injection, the pharmacokinetics and tissue distribution of curcumol liposomal injection were studied. To compare with water solution, biological half-life of liposomes injection prolonged 1.75 hours. The retention time of curcumol in each tissue of mice was prolonged. The concentration in tissues was determined by HPLC. The result showed that concentration of curcumol was highest in liver, followed by lung, spleen, kidney, brain, heart. These studies provided a basic foundation for further development and research of curcumol liposomes.
