RNA splicing is a basic element at regulation of gene expression in eukaryote.It plays an important role on organ differentiation and development in multicellular organisms.SR protein family is an important splicing factor,they have RS domain which enrich serine and arginine.Our object is CAPER belonged to SR protein family.Its homologous gene is HCC1 in human being and Rsd1 in Schizosaccharomyces pombe.Early journal indicate that Prp5,it is an ATPase,bridges the spliceosome components U1 and U2 snRNP.Then people detected Rsd1 mediate this bridge.Then people detected abnormal expression of HCC1 in kinds of cancer.It induces disorder on cell proliferation and vessel growth.Then people acquired structure of HCC1 co-crystals with splicing factor SF3B155,which is core U2 snRNP.Then detected HCC1 regulates the alternative splicing of VDGF(vascular endothelial growth factor)and calcitonin,but the detail is unknown.In addition detected HCC1 as a co-activator on transcript of estrogen hormone receptor and activator Protein1 by bridging transcript factors and machinery,then activate target gene transcripted.HCC1 bind activator Protein1 subunit c-Jun or estrogen hormone receptor,then bind ASC-2mediate transcript machinery.So far we have a lot of informations about HCC1,but a little in Drosophila melanogaster.Only acquired that CAPER join alternative splicing of Dscam ?TAF1 and signal regulation of RAS/MAPK.We study CAPER function in Drosophila melanogaster include that RNA splicing ?transcription.we use the GAL4/UAS expression system knock down CAPER expression in Drosophila melanogaster and transfect RNA into S2 cells.The results of the study are as follows:1.In S2 cells knock down CAPER expression by transfection of shRNA or dsRNA,detect alternative splicing,the results indicate that CAPER affect RNA splicing?transcript intension and terminal pre-RNA.2.Gaining whole body RNAi strain by GAL4/UAS expression system detect activator Protein1 subunit c-Jun or estrogen hormone receptor related gene,the results show that four gene are affected and Dysb protein have opposite effects.3.By GAL4/UAS expression system,detected its important role in the eyes and the central nervous system.The studies indicate that CAPER affect RNA splicing?transcription and terminal pre-RNA.On this account affect development of Drosophila melanogaster.
