Interneurons plays an important role in the process of the brain’s neural network formation,the interneurons and excitatory neurons cooperative regulating human cognition,learning and memory,other sports activities and so on.The disorder or lack of interneurons may lead to a series of neurological and psychiatric diseases,such as Alzheimer’s、Parkinsonism’s、Huntington’s disease、Epilepsy and Autism Etc.Along with the advance of Biomedical Science,the study of neurological disease pathogenesis has been updated,but the complexity and difficulty therapy of nerve disease to perplex even devoted scientific researchers.To take the advantage of cell differentiation in vitro and in vivo neural stem cell transplantation therapy is the important tool for the study of these diseases,Research finding that it have an integral connection with interneurons to occur the neurological and psychiatric disease.Therefore,It is a good foundation for the basis for studying neurodegenerative diseases more deeply that discussioning how interneurons to differentiation in vitro and using neural stem cells to repair neurodegenerative diseases.It is the main purpose of this topic that improving the directional differentiation proportion of interneurons,and respectively to discusses the following questions:(1)How the cell density impacts on the differentiation of interneurons;(2)What the time expression of interneuronal differentiation;(3)How the different concentrations of Purmorphamine influence on the differentiation proportion of interneurons;(4)How the different density of Sonic hedgehog effect on the differentiation proportion of interneurons;(5)How the different concentration combinations of PM and Shh influence on the differentiation ratio of interneurons;(6)How the XAV939 effect on the differentiation proportion of interneurons.From the whole research we get the following corresponding conclusion:It is more advantageous to the differentiation of interneurons under the density of 4000 cells per well(96-well plates);It is consistent for Interneurons differentiation happening time points in vitro with the body development time,and with the continuation of time,GABA.CR(calretinin).CB(calbindin)and PV(parvalbumin)appears in turn;It may has a certain role in promoting for interneurons differentiation under the action of PM,and it could ahead of differentiation time of interrneurons,it is a significantly higher differentiation proportion for GABA and CR under the 1.5uM/L PM;Shh could promote the differentiation of GABA、CR and PV,and it has a significantly higher for differentiation proportion of GABA and CR under the 25ng/mL Shh;At the density of lOng/mL Shh,it is a significantly higher raise for differentiation scale of PV;It has a significant impact for interneurons differentiation that The different concentration combination of the PM and Shh,at the 1 Ong/mL Shh and 1 uM PM,it could significantly highest improve the differentiation proportion of GABA;Under the luM PM and 25ng/mL Shh,the differentiation proportion of CR is highest than other groups;Compared with single factor experiment,the best combination of two-factor of PM and Shh experiment is more advantageous to the directional differentiation of interneurons;It is no significant influence on differentiation of interneurons for XAV939.
