The genus Macrotermes is one kind of fung-growing termites belonged to Macrotermitinae(Isoptera: Termitidae).Except M. barneyi and M. annandalei, other species were less attention, and need to do more research; Now because many species belonged to Macrotermes, difficultly to classification, complicated distribution,and than we cann't find more information in Chinese Macrotermes.In this paper, based on the morphological identification, 19 Macrotermes species from China was described by morphological analysis and mitochondrial COI, COII gene were amplification and sequenced than phylogenetic analysis base on COI, COII gene; The complete mitochondrial genome sequence of tow kinds of mound-mading termites, M.annandalei and M. yunnanensis located in yunnan were sequenced and annotated.1.M. chebalingensis?M. luokengensis?M. orthognathus?M. zhejiangensis and M.longimentis have closed relative with M. barneyi;2.M. menglongensis and M. guangxiensis maybe were M. annandalei;3.The molecular data exactly can't distinguish between M. denticulatus and M.jinghongensis;4. Distribution of the genus Macroitermes had expanded in China; we found M.barneyi also have distribution in guizhou and M. yunnanensis not only distribution in jinghong and Meng Lun, but also other city in yunnan province;5.The obtained sequences length of whole mitochondrial genome of M. annandalei and M. yunnanensis were 15375 bps and 15965 bps respectively.6.The mitochondrial genome of tow yunnan native Macrotermes mound-mading species contained an extra tandem repeat in their longest non-coding region.The difference between two genomes promised to serve as the markers distinguishing two species.7.The genus Macrotermes origined from M. subhyalinus and M. natalensis were at the basal position of Macrotermes within the phylogenetic tree, Africa, M. annandalci and M.yunnanensis were inferred closed relationship as a sister group, were more evolutionary than tow afric species, but M. Barneyi more evolutionary than the others.8.Multigenes NADH dehydrogenase complex among 5 species and ATP8 harbored higher mutation rate and Ka/Ks index, suggested that associated with species environment adaptability possibly.9.The mitogenomics sequeces among 5 species presented disciplinary difference,which had high relativity with its distribution and habits.The results of this study have made contribution to classification and evolutionary relationships in Macrotermes, we also want establish DNA barcode for Macrotermes species identification base on COI and COII sequences, than enrich our understanding of genus Macrotermes. At the same time, explored what mitochondrial genes may be related to species evolved to adapt to the environment of the related physiological and behavioral characteristics, and enrich the termites mitochondrial genome database.
