GNRs was prepared by seed growth methods, firstly. And then the11-mercaptoundecanoic acid(MUA) and low molecular weight polyethylenimine PEI was conjugated to the surface of GNRs to form GNRS/MUA/PEI. The GNRS/MUA/PEI were characterized by transmission electron microscopy(TEM) ?UV-vis absorption spectra and Zeta potential. In vitro cytotoxic effects of GNRS/MUA/PEI(1.8KDa) were quantified by MTT assay, the results show lower toxicity and better biological safety. The agarose gel elec-trophoresis test confirmed the complex could integrated with the si RNA effectively, and when the volume ratio of the GNRS/MUA/PEI and the siRNA was 6:1, we got the best results.The transfection efficiency was observed by the Fluorescent inverted microscope, the results showed that the FAM-siRNA was mediated into the MCF-7 cells effectively,the transfection efficiency was about 79.6%. The nano carrier of GNRS/MUA/PEI(1.8KDa) has the advantages of preparation simplely and low toxicity, can be used as a new efficient drug and gene carrier for further biological applications.
