Inter-simple sequence repeat(ISSR) markers were used to investigate the genetic diversity within and among ten natural populations of Vicia unijuga A.Br. 120 discernible DNA fragments were amplified by ten ISSR primers, 117 of them(97.5%) were polymorphic. On average each primer produced 12 bands including 11.7 polymorphic sites. On this basis we explored the correlation between genetic diversity of Vicia unijuga and bioclimatic environment factors such as altitude, rainfall, soil pH.At the species level, high genetic diversity was detected(PPB: 97.5%; Nm: 0.4553; Gst: 0.5283;H:0.2632). However, relatively low genetic diversity existed within populations, the percentage of polymorphic loci rate ranging was from 26.76 to 49.70%. Analysis of molecular variance(AMOVA)(50.51%) and Shannon's information measure(40.46%) showed a relatively high level of genetic differentiation among populations. Analysis results showed that the ten populations of Vicia unijuga had high genetic diversity, but among the populations a certain degree of genetic differentiation had occurred, the genetic differentiation between the populations was higher than within populations. At the same time, it also elucidates that no matter under low ambient pressure of Yantai or high ambient pressure of Hezuo, the Vicia unijuga have high level of diversity, but in the medium pressure regions the genetic diversity displayed lower level. We found that the genetic diversity showed a high- low- high pattern along with the increased environment pressure.The results showed that the genetic diversity of Vicia unijuga is an integrated effects of various environment factors, related to the magnitude of the environment pressures but don't have a causal relationship with particular factors. To date, almost research about Vicia unijuga focused on phenotype and physiology, in the study, we analyzed genetic diversity and genetic structure of Vicia unijuga at the molecular level, combined with environment factors and phenotypic factors analyzed the correlation between genetic diversity and environment under different microhabitat. The results of this study not only has a reference value to breeding and rational utilization of Vicia unijuga genetic resources, but also has a very important role in studying the relationship between genetic evolution and environment.
