The planthopper family Ricaniidae(Hemiptera : Auchenorrhyncha : Fulgoroidea)contains about 60 genera and 460 species, our country has recorded 5 genera and 36 species. Ricaniidae is one of the key group of system evolution. What's more, there are lots of insects as a kind of important pests of agriculture and forestry. Therefore studying Ricaniidae has an important theoretical value and economic significance. This article deeply and comprehensively studied the molecular phylogeny of Ricaniidae. This study involves DNA barcoding, Molecular identification of allied species, Corresponding relation between adults and larva based on molecular data and phylogeny based on molecular data. Results are as follows:1. Study of DNA barcoding(1) Screening out the candidate gene fragment of DNA barcodingThrough analyzing sequence composition, base substitution and genetic distance of 16 S rDNA, Cytb, CO I, 28 S rDNA, ITS 1 and ITS 2 of Ricaniidae, respectively, it is concluded that Screening mitochondrial CO I gene can be as candidate DNA barcodings of Ricaniidae.(2) verified the feasibility of candidate gene fragmentsThrough studied CO I genes of 9 samples of 3 species(Pochazia guttifera, Euricania clara and Ricania fumosa), the results showed that its genetic distance between intraspecific is 0.0349±0.0260 and interspecific is 0.1097±0.0285, it has significant difference to distinguish and identify the unknown species effectively.(3) The initial construction of DNA barcoding database of Chinese Ricaniidae.Through this study, a total of 29 mitochondrial CO I gene sequences were obtained, with 670 length consequences, included 67.2% of A+T. Initially constructed DNA barcoding database of Chinese Ricaniidae.2. Molecular identification of crptic speciesSelected 16 S rDNA and Cytb genes consequences, with P. guttifera and P. discreta of Pochazia as outgroup. Respectively Molecular identified E. clara, E. longa and E. brevicula which are similar to Euricania. Through analyzing their sequence composition, genetic distance and phylogeny, it is concluded that three allied species of Euricania can be effectively identified by 16 S rDNA and Cytb genes consequences.3. Corresponding relation between adults and larvaSelected CO I gene consequences as 13 samples of 9 species of Ricaniidae to analyze sequence composition, genetic distance and phylogeny. The results showed that there are seven kinds of wide wings of wax cicada larva and adult sequence similarity reached more than 99%; interspecific genetic distance between adult and adult is 0.139 to 0.635, but genetic distance between larva and adult is 0.004 to 0.006; Phylogenetic tree showed that all larva and adult all got together, and obtained higher test support rate. Above the results, CO I can be used to solve the problems of Corresponding relation between Ricaniidae of larva and adult.4. Phylogenetic analysis(1) Phylogenetic analysis of species levelThis paper selected Euricania and Ricanoides as representative of Ricaniidae common genus to perform phylogenetic analysis of the genus. Results are as follows:Euricania is a monophyletic group, relationship between E. brevicula and E. ocellus is closer, they are more evolved group, furthermore they are very similar in appearance as same as E. longa and E. facialis.Ricanoides is a monophyletic group, relationship between R. liboensis and R. rotundatus is closer, they are more evolved group; relationship between R. melanicus and R. pipera is closer and consistent with morphological results.(2) Phylogenetic analysis of genus levelSelected 28 S rDNA gene as Ricaniidae phylogenetic analysis of genus level, through analyzing, it is concluded that relationship between Pochazia and Euricania is closer, Ricania and Ricanoides is closer too, while monophyly of Ricanula is not supported.(3) A suggestion about establishing a new genus based on some species of RicanulaBased on the results of phylogenetic analysis of 28 S rDNA genes consequences, it is concluded that Ricanula is obviously divided into two branches, but not a monophyly, and some species were also significantly different by morphology. The results suggested that more classification, based on some species of Ricanula to establish a new genus.
