The soybean is the most important grain food and major source of edible oil.But,three soybean production bases of China are threatened by salinization in different extent.Researching the salinity-tolerant soybean species which could adapt soil salinity and take advantage of soil resource reasonably has important significance.Na~+/H~+ antiporters have been found in diferent higher plant species by botanist,which were encoded by salt tolerant gene.Na~+/H~+ antiporter in plasma membrance plays the major role in moving Na~+out cell,but also preventing Na~+ transtported into overground part of plant.The plant vacuolar Na~+/H~+ antiportor moves Na~+ into the vacuoles,adjusting Na~+/K~+ration,enhancing osmotic potential of cell to easy water-deficit resulted from high Na~+ concentrations.A large number of experiments demonstrate that Na~+/H~+antiportor could maintain plant growth under shorter salt press.According to GmsSOS1 sequences information given by NCBI,a full length cDNA of plasma membrane Na~+/H~+antiporter gene named GmsSOS1 was isolated from the soybean cultivar N23674.Software analysis showed that GmsSOS1 encodes a protein with 12 transmembrane domains and a large hydrophilic cytoplasmic tail in its C-terminal portion,which conform clinical characteristic of Na~+/H~+antiportor.The result of heterologous expression of GmsSOSl in S.cerevisiae show that GmsSOS1 protein mimick the function of NHA1 protein of S.cerevisiae.The bionformatics analysis showed that GmsSOS1 may be localized in the plasma membrane.Constructing plant expression vector pCAMBIA1305-GmsSOSl enables gene 35S::GFP-GmsSOSl expressing in cell of Tobacco.The result of sub-cellular location showed that Na~+/H~+ aniporter is membrance protein located in plasma membrane.Thus,it appears that GnmsSOS1 encodes a salinity-inducible plasma membrane Na~+/H~+ antiporter.
