The fusant of Lactobacillus acidophilus and Bacillus licheniformis owns the fine qualities of both parents, further enhance the probiotic function and ease of preparation. This study will explore the training conditions of the fusant, induce the spore to form, be microcapsuled, and further improve the gut survival rate. Results show that the characters of individual form, colony and liquid culture were obviously different from its parent strain, the fusant owns the ability to produce acid of lactobacillus acidophilus and spore generation capacity of bacillus licheniformis.The growth of fusant in the classic formula: CM> beef extract peptone?LB> MRS. The suitable initial pH is 6,7 and 5 for CM, MRS and LB medium, respectively. The initial pH of beef extract peptone medium shows no big impact on the culture of fusant, though.37? in CM medium is more advantageous to grow fusant, while the best temperature of other medium is 35 ?. Except MRS culture medium which is suitable for oxygen insulation stand cultivation, the other three kinds of culture medium are match with oxygen cradle cultivating mode.The induction ability in spore information of different solid medium:gauze> corn cob> sawdust, and was positively related. When the content is 0.1 g/ml, gauze, corn cob and sawdust induction of spore rate were 79%,71% and 63%, respectively.Treated in the artificial simulated gastric fliud ?SGF? in 2 hours, the survival rate of fusant is 0, while the survival rate of spore was 34% after 2 hours'processing, indicating that some of spore germinated into the vegetation form which is intolerant to acid environment in the watery environment and got deactivated, so microcapsule was necessary for even spore form. The cultivation function of fusant can be shown in the vegetative forms, treated in the artificial simulated intestinal fluid ?SIF? in 6 hours, the germination rate of spore was 82%, and the exist rate of spore was only 16%.When the fusant was microcapsuled, the formula of microcapsule with certain embedding rate is: 1%of sodium alginate,3% of CaCl2,0.6% of Twain-80. Without embedding, after 2 hours' treatment in SGF, the number of fusant dropped to 3.81×106 cfu·g-1 from 1.12×107 cfb·g-1, while the number of microcapsuled fusant dropped to 1.10×106from 1.80×106 with the survival rate of 61.1%, which shows that the capsule has good stability in the artificial simulated gastric fluid and has strong protection for the fusant. Intestinal soluble test results show that the number of microcapsuled fusant increased to 2.90×106 cfu·g-1 in 60 min from 4.33×104 cfu·g-1 in 20 min. It indicates embeded microcapsuled fusant can be effectively released in intestinal fluid.
