In order to explore the effect of mi RNA and high-fat diet on the fat storage hibernator,when the the changes of energy regulation mechanism and adaptability happens from the stage of lipid storage before hibernation to hibernation,Daurian Ground Squirrel was chosen the research object,we used the the second generation of transcriptome sequencing(RNA-Seq)technology to sequence the white adipose tissue of four stages: the Innitial-Fattening stage,the Rapid-Fattening stage,the Finished-Fattening stage and the Hibernation stage,and then analyzed the differentially expressed microRNAs which were related to metabolism in Experiment No.1.Main results and conclusions are as follows: 1.In fatty acid synthesis pathway,rno-mi R-30a-3p,rno-miR-30e-3p and rno-miR-106b-3p overexpressed in the Finished-Fattening significantly when the weight of Daurian Ground Squirrel is on the decline,indicated these microRNAs may inhibit fatty acid synthesis.2.Rno-miR-139-5p,rno-miR-185-5p,rno-miR-206-3p,rno-miR-23b-5p and rno-mi R-29b-2-5p were down-regulated in the Rapid-Fattening stage and the Finished-Fattening stage in the glycolysis/gluconeogenesis pathway;rno-miR-30e-3 p,rno-mi R-135a-3p and rno-194-5p were down-regulated in the Rapid-Fattening stage;rno-mi R-32-5p was down-regulated in the Finished-Fattening stage,in the Hibernation stage,rno-miR-130b-3p,rno-miR-144-5p,rno-miR-194-5p,rno-mi R-196b-5p,rno-mi R-449a-5p and rno-miR-451-5p were high-regulated.In the TCA pathway,rno-mi R-181a-5p,rno-miR-350,rno-miR-7a-5p were high-regulated in the Hibernation stage.The source of energy was food,glucose spercificly,during the fattening stages when the body weight of Daurian Ground Squirrel increases;the metabolism of glucose reduced and transformed into lipid metabolism as the energy source primarily when comes into hibernation stge.In order to explore the effect of the high-fat diet on the glucolipid metabolism of Daurian Ground Squirrel,we provided high-fat diet to Daurian Ground Squirrel that were in the Rapid-Fattening stage.The respiratory ratio and the level of blood glucose were measured before and after feeding,the body weight and energy budget were measured during feeding,hibernation array were then observed in the hibernation stge.Main results and conclusions are as follows: 1.High-fat diet feeding could increase the body weight of Daurian Ground Squirrel,delay the arrival of the Finished-Fattening stage by increasing the digestibility and metabolizable energy efficiency;2.The respiratory ratio decresed after high-fat diet feeding that indicated lipid metabolism as the energy source primarily;3.Blood glucose tolerance decreased after high-fat diet feeding,but the insulin sensitivity did not change;4.High-fat diet fedding for only one month had no effect on hibernation.
