In this study,we got ethanol extraction of Chimonanthus praecox by utilizing ultrasonic-assisted extraction with organic solvents in room temperature.After that,we got extraction of petroluem ether,ethyl acetate and butyl alcohol respectively by gradient extraction with different organic solvents one after another.We tested the antiacetylcholinesterase activity of petroleum ether extract,ethylacetate extract and butyl alcohol extract of C.praecox leaves by using improved Ellman method,the result shows that the stronggest one is ethyl acetate extraction while it has a IC50 value is 7.96 mg/mL.To discover the material basis and mechanism acetylcholinesterase inhibitory activity of C.praecox leaves furtherly,In this paper,We used some techniques like column chromatography method and sephadex gel column method to explain ethyl acetate parts of ethanol extraction from C.praecox leaves systematically with identificate the comounds subsequently.At the last we got 22 fractions last.We analysed fraction 1 with gas chromatography-mass spectrometry,14 compounds were identified,the highest content is ethyl 9,12,15-octadecatrienoate and it accounted 19.29% of fraction 1.With other fractions,we isolated eight compounds(1-8),and structures of 6 compounds were identified which named: stigmasterol(1),?-sitosterol(3),berberine(4),d-calycanthine(5),scopoletin(7)and kaempferol(8)?We utilized modified Ellman method to test the 8 compounds' acetylcholinesterase inhibitory activity.And compound 4,7 and 8 showed tremendous values while their IC50 were 4.98 mg/mL,3.48 mg/mL and 5.34 mg/mL,respectively.
