Three dimension scalar field data shows the state of a position and play an important role in topographic survey, biological medicine, weather forecast, aerospace and other fields. In the past few decades, with the rapid development of protein collection equipment and computer calculation performance, the study of proteins has become one of the most active and dynamic frontier research fields in biotechnology.Topology analysis is the key way to explore the protein data,We can effectively use of high-performance computer by establish a suitable model. However,The traditional way of topology analysis is based on discrete framework, which has some drawbacks, such as non-smoothness,low precision and hard to apply. In this paper, the main research object is the protein molecular field, and based on the model under the framework of continuous, calculating the protein molecules and analysis the field data,in addition, this article also visualize the protein topology framework and isosurface, provide a reference for researchers in protein. This paper is based on the direction of the seven box spline model and use the Morse theory topology analysis, proposes a method detecting active sites of proteins by extracting features of molecular fields of proteins and evolving their isosurfaces. In this process, the fields of proteins are regarded as three dimension scalar fields.First of all,a continuous model is built for the data field and critical points are extracted on the model. Then we use the method of isosurface evolution to quickly obtain a series of isosurfaces and observe the relationship between topological changes and critical points to determine the active sites of a protein. In the end,No.1A30 and N0.3J71 protein illustrates the effectivity of this method, and combine the space of the protein structure and the feature point, points out their results.
