In this study,a total of 81 accessions of 16 species from subgenus Soja and subgenus Glycine were selected from home and abroad.In order to obtain new nuclear gene which is suitable for phylogenentic analsys of genus Glycine,we choose the gene with no variation site was found in wild soybean and cultivated soybean by using laboratory sequencing data and the data of soybean resequencing from internet databse.In addition,combined with the selected gene sequences from the literature to construct the phylogenetic tree of soybean.The main result as below:1?In this study,we obtained 14 gene fragment in total.Four cpDNA fragments and three nDNA fragments were selected from the literature.Seven newly devoleped nuclear genes were selected and characterized by higher primer universality,moderately mutation rate(about 10%),low copy and assemble easily.2? The sequencing length of four cpDNA fragments was 2068 bp,SNP was 95,.? value was 0.00925,? value was 0.01100.The sequencing length of three nDNA fragments was 1074 bp,and the SNP was 170,? value was 0.03188,? value was0.03223.The sequencing length of seven newly devoleped nDNA fragments was2567 bp,and the SNP was 258,? value was 0.02024,? value was 0.01705.3?Analyzed the genetic diversity within different species of genus Glycine,which remains higher differences.The ? and ? values of G.can,G.cla,G.tab and G.tom are relatively high,and the ? and ? values of G.tab are significantly higher than others,which is indicate its high diversity.4?Based on phylogenetic analysis of genus Glycine,subgenus Glycine and subgenus Soja was divided into two major claed.Within subgenus Glycine,there are three main branches.These results are consequence with previous study.Analyzed cpDNA and nDNA separately,the results showed that the classification of the phylogenetic tree within subgenus Glycine are different.5?Based on different kinds of gene fragments to construct phylogenetic trees,G.tabacina were divided into two different groups.G.tab 6-G.tab 10 are from China,which have 80 chromsomes.G.tab 1-G.tab 5 are from Australia,which have 40 chromsomes.The differences might be due to the tetraploid G.tabacina(2n=80)is a large ancient polyploid complex,with high genetic diversity between the diploid G.tabacina.On the other hand,because of the correlation between geneticdifferentiation and geographic distance among populations,may be also due to long-term environmental and geographical isolation,exacerbated the genetic differentiation between them.
