AFP is a negative regulator of ABA signaling that promotes ABI5 protein degradation and weaken ABA signaling regulation,by targeting upstream of ABI5.So AFP is closely related to promote the germination of embryo and inhibit seed dormancy of regulatory mechanism.In this study,white-grained wheat varieties with different PHS tolerance were selected to amplified the T?AFP which located in chromosomes of 2A,2B and 2D.The result showed that new alleles were not detected in T?AFP-A and T?AFP-D in different PHS tolerance wheat,but two new alleles were detected in T?AFP-B,named T?AFP-Ba and T?AFP-Bb.Compared with the T?AFP-Ba and T?AFP-Bb,the T?AFP-Bb had a 4bp deletion in the 5'UTR regions.The average GI%values of T?AFP-Ba and T?AFP-Bb with different allelic variants were 30.72 and 45.18,Statistical analysis indicated two calster GI%date of T?AFP-Ba and T?AFP-Bb were significantly different(P=0.0002).So a STS marker of AFP-BF was developed.In most case,genotypes of T?AFP-Ba had more PHS resistence than genotypes of T?AFP-Bb.The allelic variation of 5'UTR in T?AFP-B affected the transcript expression level of T?AFP-B using method of RTq-PCR.The transcript expression levels of T?AFP-Ba were all more than T?AFP-Bb in seeds at 10DAP,20DAP,30DAP,40DAP,break dormancy 24h and dry seed.Besides,the transcript expression levels of T?AFP-Ba and T?AFP-Bb had a trend of increased firstly and then decreased in the different development time,and the expression level reached the highest at 20DAP and lowest at 40DAP.In addition,the dates showed that allelic variation of 5'UTR influences the stability of mRNA of T?AFP-B using mRNA inhibitors and RTq-PCR.For genotypes of T?AFP-Ba and T?AFP-Bb,mRNA had different half-time in flags,imbibed mature seeds and 20DAP seeds.The result illustrated that the date of half-time of mRNA of T?AFP-Ba is higher than the T?AFP-Bb in each type of materials.So,the stability of mRNA of T?AFP-Ba was more than T?AFP-Bb.
