Gerbera delavayi Franch.belonged to Gerbera,distributed in Yunnan province,southern Sichuan province and northern Vietnam,and grew in barren grassland of 1800?3200 meters.The dry "stone flower" leaves had good combustion,the ancestors used to them for the fire,so it was called "fireweed".G.delavayi had extensive application value.First of all,the wool fiber in the back of leaf could be the material of G.delavayi,deeply loved by the people of white,Yi,Zhuang,Lisu and Naxi,now fireweed textile technology was listed in the fourth batch of intangible culture heritage in China and Yunnan province,Sichuan province.Secondly,the roots of G.delavayi contained fungicidal ingredients,which could be used in the treatment of gastrointestinal diseases.In addition,the flower of G.delavayi had some ornamental value which could be made into fresh cut flower.Today,G.delavayi had not realized artificial planting,and the wild resources were destroyed,so the protective development of G.delavayi was imminented.Ribosomal DNA(rDNA)ITS and chloroplast DNA(cpDNA)trnL-F and rpl16 of nine populations(Pu'er,Lijiang,Eryuan,Fuyuan,Wuding,Shilin,Xinping,Kaiyuan and Dechang)of G.delavayi in southwest of China were amplified,sequenced and compared with G.jamesonii.The relationship of geographic distance and genetic distance of 9 populations was studied and NJ phylogenetic tree was constructed.At the same time,the sequences of ITS and trnL-F were studied in GenBank,and the systematic phylogenetic tree of Gerbera was constructed.The results showed that:ITS sequences analysis:(1)The whole length of ITS sequences of G.delavayi was 600?700bp,the average length was 657bp,(G+C)amounted 57.00%?57.55%.A total of 22 variable sites(ITS1,7;5.8S,2 and ITS2,3)were found;(2)The genetic distance had positive correlation with the geographic distance of G.delavayi(r~2=0.652).The genetic differentiation distance was 0.0011?0.0243,among which the genetic distance between Pu'er and other populations was the largest;(3)9 populations were divided into three branches,in which Pu'er populations was separated as the first group.The second group contained Lijiang and Eryuan provenances.The other six populations(Fuyuan,Wuding,Dechang,Shilin,Xinping,and Kaiyuan)were clustered together.According to the study of Gerbera,comparative sequence length of some Gerbera was 749bp,444 conservative sites,247 polymorphic loci,and 154 information sites.The NJ phylogenetic tree had two branches.The similarity rate was more than 90%,which proved that the genetic relationship of Gerbera is so close.trnL-F sequences analysis:(1)The whole length of trnL-F sequences of G.delavayi was 800-1000bp,the average length was 880bp,Among them,the G+C content of trnL-F sequence was lower 28.79%?36.35%;the trnL-F sequence had 241 mutation sites,the trnL(UAA)sequence had 42,the rest in the trnL-trnF sequence was 199;(2)There was no correlation between geographical distance and genetic distance(r~2=0.076),and the genetic relationship of 9 populations of G.delavayi was not good.According to the study of Gerbera,comparative sequence length of some Gerbera was 903bp,825 conservative sites,30 polymorphic loci,and 14 information sites.Most of the variation points could be used as the specific point of a species,and the relationship was fitted well.The NJ phylogenetic tree also showed two branches,The similarity rate was more than 60%,which proved that the genetic relationship of Gerbera was so close.rpl16 sequences analysis:(1)The whole length of rpl16 sequences of G.delavayi was 700?800bp,the average length was 737bp,(G+C)amounted 52.22%?52.80%.A total of 48 variable sites were found;(2)The genetic distance had positive correlation with the geographic distance of G.delavayi(r~2=0.730).(3)9 populations were divided into three branches,in which Pu'er populations was separated as the first group.The second group contained Eryuan provenances.The other seven populations(Lijiang,Fuyuan,Wuding,Dechang,Shilin,Xinping,and Kaiyuan)were clustered together.
