The Purification And Behavioral Characterization Of Mas-related G Protein-coupled Receptor X2

MrgprX2 is a member of Mas-related genes expressed in the human dorsal root ganglion and mast cells.MRGPRX2 is expressed in small neuronssensory of ganglia and mast cells.A series of cytokines and genes,such as substances P,vasoactive intestinal peptide,corticosteroids effect a lot.MRGPRX2 is associated with nociceptive stimulation,adrenal secretion,and degranulation of mast cells.Mast cells are a white blood cell.Specifically,it is a granulocyte derived from bone marrow stem cells,which is part of the immune and nervous immune system and contains many particles rich in histamine and heparin.Although the most famous function is their role in allergies and allergic reactions,mast cells also play an important role in wound healing,angiogenesis and immune tolerance.Human mast cells are activated and secreted by histone peptides such as substance P and somatostatin.MAS-related genes X2(MRGPRX2)is a member of the G protein coupled receptor superfamily.MAS-Related genes X2(MrgprX2)is a MAS-related gene(Mrgs)family Gene,located in chromosome 11 short arm of the 15 th district.In this study,the expression of the protein was expressed by Bac-to-Bac baculovirus expression system using insect cells as expression host.The initial extraction of the protein was carried out by Ni affinity chromatography and then a large number of homogeneous proteins were obtained by Superdex200 increase 5 / 150GL(GE Healthcare)to prepare for subsequent crystallization.
