In this paper,the ultrastructural and immunological characteristics of the intermediate-type fibers of three ciliates were studied by using the improved sampling method of scanning electron microscopy(SEM)and the immunofluorescence technique.Obtained sample preparation conditions of Stylonychia mytilus,Euplotes vannus,Paramecium caudatum,respectively showed about 10 nm intermediate-type filament in different parts of three ciliates by scanning electron microscopy.Then select three kinds of antibody,Anti-Cytokeratin(AE1+AE3),Anti-Lamin(A+C)and Anti-Vimentin for immunological experiments on three ciliates intermediate-type fiber.Results confirmed that different intermediate-type filament distributed in different parts of three ciliates.Specific results are as follows:(1)There're rich intermediate-type fiber in the subcutaneous of three ciliates.The diameter was 10 nm and covering the variety of organelles under surface of the membrane.Some fiber bundles into 30 nm structure.The fiber layer thickness was: Paramecium caudatum>Euplotes vannus >Stylonychia mytilus.(2)A second layer membrane structure was found in Euplotes vannus and Stylonychia mytilus for the first time.There is sill not second layer structure in Paramecium caudatum,but the fiber layer is more dense than other two ciliates.(3)The nucleus of Stylonychia mytilus appeared a hemispherical shape with many nuclear pores,was surrounded by dense intermediate-type filaments.(4)The whole cytopharynx of Stylonychia mytilus like a funnel closely packed with intermediate-type fiber and other microtubule.The end of the cytopharynx extend into a slender tube,deep into the cytoplasm.The cytopharynx of Paramecium caudatum shaped a coiled egg roll with a lot of fiber arranged in a crisscross pattern and some cilia,the end was intumescent.These results are not the same as the shape and structure of Stylonychia lemnae and Pseudourostyla cristata.In the cytoplasm,the intermediate-type filament was also found surrounded food vacuole structures.(5)Anti-Cytokeratin(AE1+AE3)has strong responses around the food vacuole of Euplotes vannus and Stylonychia mytilus.In Paramecium caudatum,it was found densely distributed around the nucleus.And they showed even distribution and net shape in the cytoplasm.(6)The distribution area of Anti-Vimentin in Stylonychia mytilus were AZM and food vacuoles.Under the dorsal and ventral surface of Euplotes vannus,the reaction zone is obviously vertical belt.In Paramecium caudatum,cytopharynx region and cortical area showed a strong reaction.(7)Anti-Lamin(A+C)only appeared a strong reaction around the nucleus of Stylonychia mytilus.But there is not any reaction in the nucleus region of Euplotes vannus or Paramecium caudatum.The cytostome area showed a number of distribution in three ciliates,especially around AZM and cytopharynx of Euplotes vannus.
