Cascade reactions are popular in industry and living organisms by which a series of chemical reactions proceed in an order that the product of one reaction is the substrate of another.Inspired by multi-enzymes systerm within cells,two types of peptides(Azo-Gly-Phe-Gly and Fmoc-FFH)were designed as the scaffold for constructing intergrated enzymes(INAzyme).The INAzyme were obtained by co-assembling peptides with hemin and glucose oxidase(GOx).The conclusions can be drawn as follows:1.Light-responsive peptide(Azo-Gly-Phe-Gly)was used to construct a light-responsive INAzyme-PepN containing hemin and GOx.The phosphate could facilitate the self-assembly of the peptide.The peptide-based nanofibers protected hemin from the formation of low-active dimer.Since the proximity effect of peptide nanofibers,the mass transfer limitation of intermediate product was suppressed,which therefore increased the catalytic efficiency on cascade reactions.Moreover,the structure and catalytic activity can be modulated through the light-responsiveness of INAzyme-PepN.2.Fmoc-FFC,Fmoc-FFR and Fmoc-FFH were design based on the amino acid composition in the active site of peroxidase.The effect of peptides on the configuration and catalytic activity of hemin was investigated.INAzyme-PepH was prepared through the co-assembly of Fmoc-FFH,hemin and GOx.The reaction efficiency of INAzyme-PepH was about 6 times that of free catalysts.Finally,INAzyme-PepH was used for the detection of glucose which displayed favorable linear relation between the concentration of glucose and the absorbance.
