ObjectiveIn the present study,Nereis(Neanthes japonica)was used as the research object.TDC2,TA,OA and ?1 adrenergic receptors were labeled by The rabbit anti-TDC2 polyclonal antibody,rabbit anti-TA polyclonal antibody,rabbit anti-OA polyclonal antibody and rabbit anti-?1 adrenergic receptor polyclonal antibody.The results provide evidence for the presence of tyramine,octopamine and norepinephrine,epinephrine,and demonstrate that tyramine,octopamine and adrenergic systems were coexisting independently in lower invertebrates.Methods1.Western blot was used to detect the expression of TDC2,TA,OA,and ?1 adrenergic receptors in the brain of Nereis(Neanthes japonica).2.The Colophony–Paraffin(CP)embedded section technique and the Streptavidin Peroxidase(SP)immunohistochemical method were used to detect the distribution of TDC2,TA,OA,and ?1 adrenergic receptors in the brain of Nereis(Neanthes japonica).ResultsTDC2,TA,OA and ?1 adrenergic receptors showed a strong positive immune response in the brain of Nereis(Neanthes japonica).1.TDC2 mainly distributed in the membrane of the cell,a small amount of TDC2 distributed in the cytosol,and had no located in nucleus.2.TA mainly distributed in the cell membrane,a small amount of distribution in the cytosol and the nucleus.3.OA all distributed in the cell membrane.4.?1 adrenergic receptors mainly located in the cell membrane,no distribution in the nucleus,However,there are four positive immunization cells were found that ?1 adrenergic receptors filled in the full cell.5.The obvious varicose-like nerve fibers of TDC2,TA,OA and ? 1 adrenergic receptors were not showd in the whole brain of Nereis(Neanthes japonica).ConclusionsThe results of this study demonstrate that tyramine,octopamine and catecholamine neurotransmitters may coexist in the brain of Nereis(Neanthes japonica)that a lower invertebrates.
